摘要
目的观察寿胎丸对去势大鼠骨组织内骨形态发生蛋白(Bone Morphogenetic Protein,BMP)和转化生长因子-β(transforming growth factor-β,TGF-β)的影响。方法 40只雌性SD大鼠随机分为假手术组、模型组、寿胎丸组和戊酸雌二醇组,行双侧卵巢切除术,寿胎丸组予寿胎丸13.4 g/kg/d灌胃,戊酸雌二醇组予补佳乐0.104 mg/kg/d灌胃,假手术组与模型组予等量生理盐水灌胃。于术后12周处死动物,切取大鼠股骨标本分别检测股骨近端骨组织形态学、骨组织中BMP和TGF-β含量及相关调控基因的表达。结果寿胎丸可明显改善卵巢切除所引起的骨组织微观结构的破坏。模型组骨组织中BMP2、BMP4和TGF-β含量明显低于假手术组(P<0.05);与模型组相比,寿胎丸组BMP2、BMP4和TGF-β,戊酸雌二醇组BMP2的含量均明显增加(P<0.05)。同时,模型组骨组织中,BMP2、BMP4和TGF-β的基因表达明显降低(P<0.05);与模型组相比寿胎丸组BMP2、BMP4和TGF-β,戊酸雌二醇组BMP2相关调控基因的表达均明显增加(P<0.05)。结论寿胎丸可明显改善卵巢切除引起的骨组织微观结构的破化,这可能与寿胎丸提高骨组织内BMP2、BMP4和TGF-β蛋白含量和相关调控基因的表达有关。
Objective To observe effects of Shoutai pill on expression of BMP and TGF-β in ovariectomized rat. Methods Totally 40 female healthy SD rats were divided into sham group, ovariectomized group, Shoutai pill group and Progynova group. The bilateral oophorectomy was conducted. The rats in Showtai pill group were intervened by Shoutai pill with the dosages 13.4 g/kg/d and the rats in Progynova group were intervened by Progynova with the dosage of 0.104 mg/kg/d. In Sham group and ovariectomized group, equal quantity saline were intragastric administration. The rats were executed at 12 weeks after the operation, the histomorphology changes of bone were observed with HE stain, the content of BMP2, BMP4, BMP6, BMP7, TGF-β and the related regulating gene were detected. Results The application of Showtai pill could significantly ameliorate the microstructure changes of bone tissue. In bone tissues of ovariectomized group, the content of BMP2, BMP4 and TGF-β were significantly decreased than that those of the sham group( P0.05). Comparing with ovariectomized group, the content of BMP2, BMP4 and TGF-β, and BMP2 in Progynova group were significantly increased( P〈0.05).Meanwhile, comparing with the sham group, the m RNA expression of BMP2, BMP4 and TGF-β in the ovariectomized group were significantly decreased( P〈0.05). Comparing with the ovariectomized group, the m RNA expression of BMP2, BMP4 and TGF- βin of the Shoutai pill group and BMP2 in the Progynova group were significantly increased( P0.05). Conclusion The application of Showtai pill could significantly ameliorate the microstructure changes of bone tissue. This effect was likely be caused by increasing the content of BMP2, BMP4, TGF-β and the related m RNA expression.
