摘要
目的探讨补肾健脾、解毒利咽法治疗Ig A肾病(Ig AN)的作用机制。方法将72只Wistar大鼠随机分为空白组,模型组,洛丁新组,补肾健脾、解毒利咽法高、中、低剂量组(简称为中药高、中、低剂量组)。采用口服BSA和尾静脉注射LPS方法建立Ig AN大鼠模型。观察给药8周后大鼠各时相尿红细胞计数、24h尿蛋白定量(UTP)及肾组织基质金属蛋白酶-9(MMP-9)、金属蛋白酶组织抑制剂-1(TIMP-1)的含量变化。结果1)与模型组相比,各组大鼠24 h UTP均有所下降,其中中药中剂量组下降最为显著(P<0.05);2)与模型组相比,中药各治疗组尿红细胞计数明显减少(P<0.05),以中药中剂量组减少显著(P<0.01);洛丁新组尿中红细胞计数无下降(P>0.05)。3)与空白组相比,模型组大鼠肾组织中MMP-9含量明显降低,TIMP-1含量升高,MMP-9/TIMP-1下降(P<0.05);4)给药后,与模型组相比,治疗组MMP-9含量明显升高、TIMP-1含量明显降低,MMP-9/TIMP-1明显升高(P<0.05),以中药中剂量组最为显著(P<0.01)。结论补肾健脾、解毒利咽法能降低Ig AN模型大鼠血尿、蛋白尿,升高肾组织中MMP-9含量,降低TIMP-1含量,使MMP-9/TIMP-1趋于平衡,疗效优于洛丁新组。
Objective To study the effect and mechanism of Bushenjianpi and Jieduliyan method on Ig A nephropathy. Methods The 72 Wistar rats were randomly divided into blank group,model group,Lotensin group,Bushenjianpi and Jieduliyan high,medium and low dose group（ is called traditional Chinese medicine high,and low dose group for short）. The Ig A nephropathy model rat were by the method of feeding with bovine serum albumin（ BSA）and injecting tail vein with lipopolysaccharide（ LPS）. Eight weeks administration later,each phase＇s urine red blood cell count,UTP,Serum and kidney tissues of MMP-9 and TIMP-1 content were observed. Results 1） Compared with model group,every groups＇ UTP decreased and traditional Chinese medicine group decreased most（ P〈0. 05）. 2）Compared with model group,urine red blood cell count decreased significantly of every traditional Chinese medicine group（ P〈0. 05）,especially traditional Chinese medicine medium dose group improved significantly（ P〈0. 01）,but Lotensin group urine red blood cell count no drop（ P〈0. 05）. 3） Compared with blank group,MMP-9 on model group＇s kidney tissue decreased while TIMP-1 content rose,and MMP-9 / TIMP-1 decreased（ P〈0. 05）. 4） After administration,treatment groups MMP-9 content rose significantly,TIMP-1 content decreased significantly,and MMP-9 /TIMP-1 rose significantly（ P〈0. 05）,traditional Chinese medicine medium dose group＇s data is the most significant（ P〈0. 01）. Conclusion Bushenjianpi and Jieduliyan method can reduce the model rats hematuria and proteinuria of Ig A nephropathy,and it can raise MMP-9 content in kidney tissue,reduce TIMP-1 content,and balance MMP-9 /TIMP-1 which is better than Lotensin.
