摘要
目的观察益肾汤治疗对IgA肾病模型小鼠肾组织中MMP-9、TIMP-1表达水平的影响。方法将选择8~10周龄SPF级雄性小鼠,随机将100只小鼠分为对照组和实验组,对照组20只,对小鼠给予常规喂养,正常饮食、饮水,自由活动;实验组80只,在对照组的基础上,对小鼠进行益肾汤灌胃。观察比较五组小鼠的毛发、体质量、尿量及排尿颜色。观察并记录各阶段小鼠尿红细胞计数、24 h尿蛋白定量(24 h urinary protein quantification,UTP)、肾组织基质金属蛋白酶-9(Matrix metalloproteinase-9,MMP-9)及金属蛋白酶组织抑制剂-1(Tissue inhibitor of metalloproteinase-1,TIMP-1)的含量变化情况。结果模型组小鼠的MMP-9含量较对照组显著降低,TIMP-1含量显著升高,MMP-9/TIMP-1<1;益肾汤低浓度组、中浓度组和高浓度组小鼠MMP-9含量较模型组显著升高,TIMP-1含量显著降低,MMP-9/TIMP-1显著升高,其中,中浓度组的疗效更显著,差异存在统计学意义(P<0.05)。结论益肾汤可促进小鼠肾组织MMP-9的表达,抑制TIMP-1的表达,有效延缓IgA肾病的进展。
Objective To observe and analyze the effect of Yishen decoction on the expression of MMP-9 and TIMP-1 in the kidney tissue of mice with IgA nephropathy.Methods 100 SPF male mice aged 8~10 weeks were randomly divided into control group and experimental group.20 mice in the control group were given routine feeding,normal diet,drinking water and free activity.80 mice in the experimental group were given Yishen decoction by gavage on the basis of the control group.The hair,constitution,urine volume and micturition color of the five groups were observed and compared.The changes of urinary erythrocyte count,24-h urinary protein(UTP),matrix metalloproteinase-9(MMP-9)and tissue inhibitor of metalloproteinase-1(TIMP-1)in renal tissue were observed and recorded Results The content of MMP-9 in the model group was significantly lower than that in the control group,and the content of TIMP-1 was significantly increased,MMP-9/TIMP-1<1.MMP in the low-concentration,middle-concentration and high-concentration groups of Yishen decoction,compared with the model group,the content of-9 was significantly increased,the content of TIMP-1 was significantly decreased,and the content of MMP-9/TIMP-1 was significantly increased.Among them,the therapeutic effect of the medium concentration group was more significant,and the difference was statistically significant(P<0.05).Conclusion Yishen decoction can promote the expression of MMP-9 and inhibit the expression of TIMP-1 in mouse kidney tissues,effectively delaying the progression of IgA nephropathy.
