烫骨碎补中总黄酮及柚皮苷的含量测定 被引量:5

摘要 目的建立测定烫骨碎补中总黄酮含量的紫外可见分光光度(UV)法及柚皮苷含量的高效液相色谱(HPLC)法。方法 UV法检测波长为283 nm。HPLC法色谱柱为ODS2 C18柱(150 mm×4.6 mm,5μm),流动相为甲醇-醋酸-水(25∶4∶75),流速为1 m L/min,检测波长为283 nm,柱温为30℃,进样量为10μL。结果骨碎补中总黄酮最大吸收波长为283 nm,质量浓度在0.002 82~0.112 8 g/L范围内与吸光度线性关系良好;柚皮苷质量浓度在1.128~112.8 g/L范围内与峰面积线性关系良好,加样回收率为98.38%,RSD为1.88(n=9)。结论所建立的方法操作简单、结果准确、重复性较好,可用于烫骨碎补中总黄酮及柚皮苷的含量测定。 Objective To establish an HPLC method for the determination of total flavonoids and naringin in hot Rhizoma Drynariae.Methods UV assay wavelength was 283 nm, HPLC column was ODS2 C18 column( 150 mm × 4. 6 mm, 5 μm), mobile phase was methanol- acetic acid- water( 25 ∶ 4 ∶ 75), the flow rate was 1 m L / min, the detection wavelength was 283 nm, the column temperature was 30 ℃,the injection volume was 10 μL. Results The absorption wavelength of total flavonoids in Drynaria maximum was 283 nm,and the mass concentration in the range of 0. 002 82- 0. 112 8 g / L had a good linear relationship with absorbance; naringin concentration in the range of 1. 128- 112. 8 g / L had good linear relations with the peak area, and the recovery rate was 99. 38%, RSD was1. 88%( n = 9). Conclusion The method is simple, accurate, and reproducible and can be used in the content determination of total flavonoids and naringin in hot Rhizoma Drynariae.
