摘要
目的:考察煎煮时间及配伍对不同剂量附子煎煮液中生物碱含量的影响,阐述配伍对附子"增效减毒"的科学性,不同剂量附子煎煮液中毒性大小与煎煮时间的相关性,探索超大剂量煎煮液临床应用的安全性。方法:采用高效液相色谱法测定不同剂量、不同煎煮时间附子单煎液和复方煎煮液中6种生物碱成分含量,比较其毒性成分和有效成分溶出度。结果:5-90g黑顺片煎煮1h后,双酯型生物碱含量均完全消失,随着煎煮时间延长,煎煮液中单酯型生物碱的含量逐渐降低,45g黑顺片煎煮3.5h后,其含量逐渐消失;不同剂量单酯型生物碱溶出率有所差异,随着剂量的增加溶出率逐渐降低,总溶出量则与其剂量大小呈正相关,15g黑顺片煎煮液中溶出率为58.92%,溶出量为12.3270mg,90g黑顺片煎煮液中溶出率为43.43%,溶出量为50.513mg,两种溶出量比较,90g剂量组是15g剂量组的4.22倍;与单煎煮比较,相同剂量四逆汤煎煮液中单酯型生物碱溶出率和溶出量均有所增加,15g四逆汤煎煮液中溶出率为62.85%,溶出量为13.1490mg。结论:超大剂量附子煎煮液中生物碱溶出量增加,通过延长煎煮时间和配伍可以减低毒性,为了保证药物安全性和有效性,45g黑顺片应煎煮2.5h。
Objective：To investigate the effects of decocting time and compatibility on the content of the alkaloid of decocting water with different doses of Radix Aconiti Lateralis Preparata,to elaborate the scientificity in the effect of compatibility on reducing toxicity and synergy of Radix Aconiti Lateralis Preparata,to research the correlation of the toxic size of the decocting water and the decocting time,and to explore the security for the application of a large dose of Radix Aconiti Lateralis Preparata.Methods：The six kinds of alkaloids in single and compound decocting water with different doses and decocting time of Radix Aconiti Lateralis Preparata were determined by HPLC.Then,the toxic components and the active ingredient dissolution were compared.Results：The content of diester-type alkaloids in decocting water with 5g to 90 g of Radix Aconiti Lateralis Preparata was completely disappeared after decocting for 1h.With the increasing of decocting time,the content of mono-ester alkaloids was gradually decreased,and that in decocting water with 45 g of Radix Aconiti Lateralis Preparata gradually disappeared after decocting for 3.5h.There was difference in the dissolution rate of mono-ester alkaloids among different dose of Radix Aconiti Lateralis Preparata.With the increasing of dose,the dissolution rate of mono-ester alkaloids was gradually decreased.But,the total dissolution quantity was positively associated with the dose of Radix Aconiti Lateralis Preparata.The dissolution rate of decocting water with 15 g of Radix Aconiti Lateralis Preparata was 58.92%,and the total dissolution quantity of that reached 12.3270 mg.The dissolution rate of decocting water with 90 g of Radix Aconiti Lateralis Preparata was 43.43%,and the total dissolution quantity of that reached 50.513 mg.The total dissolution quantity of 90 g group was 4.22 times than that of 15 g group.Compared with the single decoction,the dissolution rate of mono-ester alkaloids in the same dose Sini decoction was increased,as well as the dissolution qua
