玉屏风散加味方干预COPD气道重塑相关细胞因子作用研究 被引量:14

摘要 目的探讨玉屏风散加味方对慢性阻塞性肺疾病(COPD)气道重塑大鼠基质金属蛋白酶-9(MMP-9)、基质金属蛋白酶抑制剂-1(TIMP-1)和转化生长因子β1(TGF-β1)的调节作用。方法将60只SD大鼠随机分为正常组、模型组、玉屏风散加味方高剂量组、玉屏风散加味方中剂量组、玉屏风散加味方低剂量组、阳性组,每组10只。除正常组外,其余各组大鼠采用被动吸烟加气管内注入LPS的方法复制COPD大鼠模型,玉屏风散加味方各组给予相应剂量玉屏风散加味方灌胃,阳性组给予罗红霉素胶囊溶液灌胃,正常组、模型组给予等量生理盐水灌胃。灌胃30 d后用ELISA方法测定大鼠肺泡灌洗液中MMP-9、TIMP-1、TGF-β1水平,计算MMP-9/TIMP-1比值。结果模型组MMP-9、TIMP-1、TGF-β1表达水平均明显高于正常组(P均<0.05),MMP-9/TIMP-1比值明显低于正常组(P<0.05);玉屏风散加味方高、中剂量组及阳性组MMP-9、TIMP-1、TGF-β1表达水平均明显低于模型组(P均<0.05),玉屏风散加味方高剂量组MMP-9、TIMP-1、TGF-β1表达水平均明显低于玉屏风散加味方低剂量组(P均<0.05)。结论玉屏风散加味方可延缓COPD气道重塑病变,其干预COPD气道重塑的作用可能是通过调节气道局部细胞因子MMP-9、TIMP-1和TGF-β1表达实现的,且以高剂量效果最为明显。 Objective It is to explore the regulating effect of Yupingfeng powder on MMP - 9, TIMP - 1 and TGF - β1 in COPD rats with airway remodeling. Methods 60 SD rats were randomly divided into normal group, model group, high, medium and low dose of Yupingfeng powder groups, positive group, each group had 10 rats. All the rats except in normal group were given passive smoking with LPS injected in airway to establish COPD model. The Yupingfeng powder groups were given Yupingfeng powder at the respective dose by gavage, the positive group was given erythromyein capsule solution by gavage, normal group and model group was given normal saline by garage. After treating for 30 days, the levels of MMP - 9, TIMP - 1 , TGF - β1 in bronchoalveolar lavage fluid of the rats were determined by ELISA, and MMP - 9/TIMP - 1 were calculated. Results The levels of MMP -9, TIMP - 1, TGF -β1 in model group were significantly higher while MMP -9/TIMP - 1 were lower than that in normal group (P 〈 0.05 ) , the levels in high and medium dose of Yupingfeng power groups and positive group were significantly lower than that in model group (P 〈 0.05 ) , the levels in high dose of Yupingfeng power group were lower than that in lower dose of Yupingfeng power group(P 〈 0.05 ). Conclusion Yupingfeng powder can delay the development of airway remodeling induced by COPD, and the intervention mechanism may be achieved by regulating airway remodeling related cytokines MMP -9, TIMP - 1, TGF - β1 , and the effect is most significant in high dose group.
