GLP-1受体表达与COPD气道平滑肌增殖的相关性研究

Analysis of the correlation between GLP-1 receptor expression and airway smooth muscle proliferation in COPD

摘要 目的探讨胰高血糖素样肽-1(GLP-1)蛋白表达与慢性阻塞性肺疾病(COPD)气道平滑肌增殖的相关性。方法选择2018年1月-2019年7月于喀什地区第一人民医院心胸外科行肺部手术患者为研究对象,有COPD病史并需要行肺减容术的患者为病例组(n=18),无COPD病史、肺良性肿瘤并且需要行肺部手术的患者为对照组(n=15)。检测病例组和对照组肺组织中GLP-1蛋白表达量、GLP-1(mRNA)表达量及GLP-1基因甲基化情况。结果病例组血清白细胞介素(IL)-6水平(7.304±0.137)pg/mL高于对照组(3.033±0.103)pg/mL,血清IL-8水平(0.833±0.052)ng/mL高于对照组(0.168±0.017)ng/mL,差异均有统计学意义(F=1.910、15.324,P均<0.05)。病例组肺组织中GLP-1(mRNA)表达量(0.568±0.044)低于对照组(0.912±0.145),GLP-1蛋白表达量(0.596±0.131)低于对照组(1.000±0.000),差异均有统计学意义(F=6.092、10.507,P=0.025、0.032)。病例组甲基化阳性占比(66.67%),高于对照组(33.33%),差异无统计学意义(P>0.05)。GLP-1甲基化阳性率与管壁平均厚度(WT)、管壁平均直径(WD)成正相关(β=0.140、2.167),差异均有统计学意义(P均<0.05)。GLP-1(mRNA)与WT、WD成负相关(β=-0.321、-2.609),GLP-1蛋白表达量与WT、WD成负相关(β=-0.228、-3.706),差异均有统计学意义(P均<0.05)。结论GLP-1蛋白表达与COPD气道平滑肌增殖存在相关性,可能参与了COPD的发生发展。 Objective To investigate the relationship between the expression of glucagon like peptide-1(GLP-1)protein and the proliferation of airway smooth muscle in chronic obstructive pulmonary disease(COPD).Methods Patients who underwent lung surgery in the Department of Cardiothoracic Surgery of the First People's Hospital of Kashgar from January 2018 to July 2019 were selected as the research object.Patients with COPD history and needed lung volume reduction surgery were selected as the case group(n=18),and patients with benign lung tumors without COPD history who needed lung surgery were selected as the control group(n=15).The expression of GLP-1 protein,GLP-1(mRNA)and GLP-1 gene methylation in lung tissues of case group and control group were detected.Results The level of serum interleukin(IL)-6 in the case group(7.304±0.137)pg/mL was significantly higher than that in the control group(3.033±0.103)pg/mL,the level of serum IL-8 in patients(0.833±0.052)ng/mL was higher than that in the control group(0.168±0.017)ng/mL,the differences were statistically significant(F=1.910,15.324,all P<0.05).The relative expression level of GLP-1(mRNA)in the lung tissue of patients in the case group(0.568±0.044)was significantly lower than that in the control group(0.912±0.145);the relative expression level of GLP-1 protein in the lung tissue of the case group(0.596±0.131)was significantly lower than that of the control group(1.000±0.000);the differences were statistically significant(F=6.092,10.507,P=0.025,0.032).The proportion of methylation positive in the case group(66.67%)was higher than that in the control group(33.33%),but the difference was not statistically significant(P>0.05).The positive rate of GLP-1 methylation was positively correlated with average thickness of wall(WT)and mean diameter of pipe wall(WD)(β=0.140,2.167)(all P<0.05).GLP-1(mRNA)was negatively correlated with WT and WD(β=-0.321,-2.609),the expression of GLP-1 protein was negatively correlated with WT and WD(β=-0.228,-3.706),the differences were statis
