摘要
目的探讨溶酶体相关4次跨膜蛋白质B(LAPTM4B)基因在良恶性前列腺组织中的表达差异及异常表达的LAPTM4B与前列腺癌的关系。方法采用已知微阵列(Microarray)数据分析LAPTM4B在良恶性前列腺组织中的表达差异。利用免疫组织化学技术对100例前列腺癌和80例良性前列腺标本中LAPTM4B的蛋白表达水平进行分析。结果LAPTM4B的DNA水平在良恶性前列腺组织中差异有统计学意义(P〈0.01);转移性前列腺中LAPTM4B的DNA水平明显高于原发前列腺癌(P〈0.01)。LAPTM4B的mRNA水平在高Gleason评分患者中明显增高(P〈0.05)。LAPTM4B的蛋白表达在良恶性前列腺组织中差异有统计学意义(P〈0.01)。在不同水平前列腺特异抗原(PSA)前列腺癌患者中LAPTM4B蛋白表达差异有统计学意义(P〈0.01);在不同Gleason评分的前列腺癌患者间LAPTM4B蛋白表达差异有统计学意义(P〈0.01);LAPTM4B蛋白表达在PSA生化复发的前列腺癌患者中明显增高(P〈0.05)。结论在前列腺癌组织中LAPTM4B基因的表达异常,上调的LAPTM4B与前列腺癌具有相关性,可能为潜在的前列腺癌标志物。
Objective To analyze the expression of lysosome - associated protein transmembrane 4 beta (LAPTM4B) in prostate tissues and investigate the relationship with clinicopathological variables in prostate cancer. Methods Use available microarray datasets to test the expression of LAPTM4B in pros- tate tissues. In 100 prostate cancer (PCa) and 80 benign prostatic hyperplasia (BPH) samples, we per- formed immunohistochemical staining to validate expression of LAPTM4B. Results Microarray data indi- cated that that LAPTM4B DNA copy number was significantly increased in PCa tissues relative to normal prostate tissues (P 〈 0. 01 ). Similarly, increased LAPTM4B DNA copy number was found in metastatic PCa (P 〈 0. 01 ). mRNA level of LAPTM4B is significantly over- expressed in patients with higher Glea- son score ( P 〈 0. 05 ). LAPTM4B protein was overexpressed in PCa tissues compared with the BPH ( P 〈 0. 01 ). LAPTM4B protein expression was significantly upregulated in PCa patients with, higher preopera- tive prostate specific antigen (PSA, P 〈 0.01 ) ; in patients of higher Gleason score LAPTM4B protein was significantly increased (P 〈 0. 01 ) , and in patients with BCR LAPTM4B protein expression was significant- ly higher (P 〈 0.05 ). Conclusion Overexpressed LAPTM4B is associated with some PCa clinicopatho- logical variables and is a potential PCa molecular marker.
