摘要
目的分析乳腺癌癌组织中新肿瘤相关基因溶酶体相关4次跨膜蛋白B(lysosome-associated protein transmembrane-4 beta,LAPTM4B)的表达水平及临床意义。方法研究对象为2019年1月-2020年3月在本院接受乳腺癌手术治疗的患者;应用RT-PCR法检测乳腺癌组织及癌旁组织中LAPTM4B mRNA水平,比较乳腺癌组织、癌旁组织中LAPTM4B mRNA水平及不同临床特征的乳腺癌患者癌组织LAPTM4B mRNA水平,Logistic回归分析乳腺癌临床特征与LAPTM4B mRNA水平的关系。结果乳腺癌癌组织中LAPTM4B mRNA水平显著高于癌旁组织;将乳腺癌患者癌组织LAPTM4B mRNA水平按中位数分高表达组(≥0.89,n=52)、低表达组(<0.89,n=35),导管浸润性、淋巴结转移、TNM分期Ⅲ~Ⅳ期、Ki-67阳性的乳腺癌患者癌组织LAPTM4B mRNA高表达比例分别显著高于非导管浸润性、无淋巴结转移、TNM分期Ⅰ~Ⅱ期、Ki-67阴性患者(P<0.05);Logistic回归分析显示乳腺癌病理分型、淋巴结转移、TNM分期及Ki-67表达均是LAPTM4B mRNA表达的独立影响因素。结论乳腺癌癌组织中新肿瘤相关基因LAPTM4B mRNA存在高表达现象,并与乳腺癌病理分型、淋巴结转移、TNM分期及Ki-67表达存在密切关联。
Objective To analyze the expression level and clinical significance of a novel tumor-related gene lysosome-associated protein transmembrane-4 beta(LAPTM4B)in breast cancer tissues.Methods Patients who underwent breast cancer surgery in the hospital from January 2019 to March 2020 were selected as subjects.The RT-PCR method was used to detect the LAPTM4B mRNA level,and LAPTM4B mRNA levels in breast cancer tissues and adjacent tissues and in breast cancer patients with different clinical characteristics were compared.Logistic regression analysis of the relationship between clinical characteristics of breast cancer and the LAPTM4B mRNA level was performed.Results The LAPTM4B mRNA level was significantly higher in breast cancer tissues than in adjacent tissues.According to the LAPTM4B mRNA level in cancer tissues,breast cancer patients were divided into high expression group(≥0.89,n=52)and low expression group(<0.89,n=35).The high expression ratio of LAPTM4B mRNA was significantly higher in breast cancer patients with ductal infiltration,lymph node metastasis,TNM stage III-IV and Ki-67-positive ones than in those without ductal infiltration,without lymph node metastasis,with TNM stageⅠ~Ⅱand Ki-67 negative ones(P<0.05).Logistic regression analysis showed that pathological type of breast cancer,lymph node metastasis,TNM stage and Ki-67 expression were independent influencing factors of LAPTM4B mRNA expression.Conclusion The novel tumor-related gene LAPTM4B mRNAis highly expressed in breast cancer tissues,which is closely related to the pathological type of breast cancer,lymph node metastasis,TNM staging and Ki-67 expression.
