摘要
以马铃薯渣酶解浓缩液为碳源,黄色短杆菌为发酵菌株,利用响应面法,分析了总糖(马铃薯渣酶解浓缩液添加量)、硫酸铵和玉米浆优化培养基的最佳组成,进而研究了生物素和二甲基亚砜的最佳用量。最终确定最佳发酵培养基:总糖125.81 g/L,(NH4)2SO427.20 g/L,玉米浆18.17 m L/L,生物素180μg/L,二甲基亚砜18 m L/L,且在发酵18 h时添加二甲基亚砜较佳,L-赖氨酸产量达到64.99 g/L,说明马铃薯渣可以作为发酵L-赖氨酸的原料。
In this study,fermentation of L-lysine by Brevibacterium flavum with condensed enzymatic hydrolysate of potato pomace as carbon source was investigated.The composition of total sugar(the amount of condensed enzymatic hydrolysate of potato pomace),ammonium sulfate and corn steep liquor of the medium was optimized with response surface methodology.The optimal dosage of biotin and dimethyl sulfoxide(DMSO) was further analyzed based on the optimized medium.The final optimum fermentation condition was as follows:total sugar 125.81 g/L,(NH4)2SO427.20 g/L,corn steep liquor 18.17 m L/L,biotin 180 μg/L,DMSO 18 m L/L.The DMSO was added after 18 h fermentation.The production of lysine reached 68.19 g/L and was 41.21% higher than that of non-optimal culture.The potato pomace was suitable carbon source for L-lysine fermentation.
