高脂饮食大鼠脂肪肝脂质代谢轴的变化及中药对其的干预作用 被引量:6

Changes of fatty acid synthesis pathway in rats with fatty liver disease induced by high-fat diet and the Chinese herbal decoction intervention

摘要 目的探讨腺苷酸活化的蛋白激酶(AMPK)-乙酰辅酶A羧化酶(ACCase)-丙二酰辅酶A(Malonyl Co A)脂质代谢轴在脂肪肝病理机制中的意义。方法采用大鼠脂肪肝模型,随机分正常组、模型组和中药干预组各8只。造模12周,中药干预组在第8周起按1 ml/100 g鼠重予中药(中药"HJJB"复方:红景天苷、绞股蓝总苷、姜黄素、白术多糖)灌胃治疗。观察项目:1肝组织HE染色;2生化法检测血清及肝脏甘油三酯(TG)含量,酶免法检测肝脏游离脂肪酸(FFA)、AMPK、ACCase、Malonyl CoA含量;3肝脏TG、FFA、AMPK、ACCase、Malonyl CoA含量间的相关性分析。结果 1模型组肝脏出现严重的脂肪变性,肝脏TG、FFA、ACCase、Malonyl CoA含量较正常组均显著升高(P<0.01),肝脏AMPK含量较正常组显著降低(P<0.01);中药干预组的肝脏TG、FFA、ACCase、Malonyl CoA含量均显著低于模型组(P<0.01),肝脏AMPK含量则显著高于模型组(P<0.01)。2肝脏TG、FFA含量与肝脏ACCase、Malonyl CoA含量之间均呈显著正相关(P<0.001),而与肝脏AMPK含量则呈显著负相关(P<0.001)。结论脂肪肝大鼠AMPK-ACCase-Malonyl CoA脂质代谢轴明显异常,在脂肪肝病理机制中有重要意义。 Objective To investigate the role of AMP-activated protein kinase( AMPK),acetyl Co A carboxylase( ACCase) and Malonyl Co A in the pathological mechanisms of fatty liver. Methods The rat were randomly divided into three groups. The normal group did not received the high-fat diet,the model group was induced by high-fat diet,and the intervention group was induced by high-fat diet and received Chinese herbal decoction intervention,i. e. the traditional Chinese medicine"HJJB"compound with the dose of 1 ml /100 g weight at the beginning of the eight week,there were 8 rats in each group. The observing items including: ①Histopathological examination of liver sections( H. E. staining); ②serum and hepatic triglyceride( TG) levels by biochemical method; hepatic free fatty acid( FFA),AMPK,ACCase and Malonyl-Co A by enzyme linked immunosorbent assay; ③correlation analysis among hepatic AMPK,ACCase,malonyl-Co A,hepatic TG and FFA contents. Results ①Serious hepatic steatosis were observed in the model group. The hepatic TG,FFA,ACCase and Malonyl Co A levels of the model group were significantly higher than those of the normal group( P〈0. 01). The hepatic AMPK content of model group was significantly lower than that of the normal group( t = 6. 63). The hepatic TG,FFA,ACCase and Malonyl Co A levels of the Chinese herbal decoction intervention group were significantly lower than those of the model group( P〈0. 01),and the hepatic AMPK level was significantly higher than that of the model group( P〈0. 01). ②The positive correlation was observed between hepatic TG,FFA content and hepatic ACCase and malonylCo A levels( P〈0. 001). While there were significant negative correlation between hepatic TG,FFA levels and hepatic AMPK level( P〈0. 001). Conclusion AMPK-ACCase-Malonyl Co A is obviously abnormal in fatty liver rats which probably play very important roles in pathological mechanisms of fatty liver disease.
