Determination of xylo-oligosaccharides content in health foods by high performance liquid chromatography with evaporative light scattering detector

摘要 目的建立高效液相色谱仪-蒸发光散射检测器测定保健食品中低聚木糖含量的方法。方法样品经乙醇溶液沉淀大分子多糖及高聚合度木糖,溶液经沸水浴中硫酸水解后,用氢氧化钠溶液调节p H值至中性,以乙腈-水(80∶20,V/V)为流动相,经高效液相色谱仪-蒸发光散射检测器测定样品中原有木糖的质量含量和样品中低聚木糖经硫酸水解后的木糖的含量,二者之差除以平均转换系数即得到样品中低聚木糖的含量。结果被测组分的浓度(0.054 5 mg/ml^1.09 mg/ml)与峰面积的线性关系良好(r=0.999 7),相对标准偏差(RSD)为3.03%(n=6),加标回收率为90.9%~94.6%和90.8%~95.1%。结论本方法简便、快速,适用范围广,准确度和精密度良好,可用于日常保健食品中低聚木糖的检测。 Objective To develop a method for the determination of xylo- oligosaccharides in health foods by high performance liquid chromatography( HPLC) with evaporative light scattering detector. Methods Precipitate the high degree of polymerization of xylose and macromolecular polysaccharide of the sample by the mixture of ethanol- water; then hydrolyze with sulfuric acid solution in boiling water bath; neutralized with sodium hydroxide solution; separated with a mixture of acetonitrile- water( 80∶20,V/V) as mobile phase; to determine the original quality and content of xylose in the sample and the content of xylose after hydrolysis of oligosaccharides in the sample by HPLC with evaporative light scattering detector,so as to get the content of xylose when the difference of the two formers was divided by the average conversion coefficient in the sample. Results There was a good linearity of the calibration curve between the peak area and the mass concentration of xylose in the range of0. 054 5 mg / ml- 1. 09 mg / ml( r = 0. 999 7); the relative standard deviation( RSD) was 3. 03%( n = 6); the recoveries were among 90. 9%- 94. 6% and 90. 8%- 95. 1%. Conclusion This method is found to be simple,rapid,precise and accurate.Therefore,the method can be successfully used for the determination of xylo- oligosaccharides in health foods.
