摘要
目的探讨缺氧诱导因子(HIF)-1α、血管内皮细胞生长因子(VEGF)在垂体腺瘤中的表达及其意义。方法 42例垂体腺瘤患者按病理改变分为侵袭组27例,非侵袭组15例。应用RT-PCR检测垂体腺瘤HIF-1α和VEGF mRNA的表达,用免疫组化染色检测垂体腺瘤HIF-1α和VEGF蛋白的表达,并分析HIF-1α和VEGF表达之间的相关性。结果侵袭组与非侵袭组垂体腺瘤HIF-1αmRNA表达水平比较,差异无统计学意义(P>0.05);侵袭组垂体腺瘤VEGF mRNA的表达水平比非侵袭组显著增高(P<0.05);侵袭组垂体腺瘤HIF-1α和VEGF蛋白的表达程度均高于非侵袭组(均P<0.05),并且HIF-1α、VEGF蛋白和mRNA的表达水平均呈正相关(r=0.436,r=0.890,均P<0.05)。结论 HIF-1α和VEGF蛋白的表达程度与垂体腺瘤的侵袭性有关。
Object To explore the expressions and their significance of the hypoxia-inducible factor( HIF)-1α and vascular endothelial growth factor( VEGF) in pituitary adenomas.Methods The mRNA expressions of HIF-1α and VEGF were determined by reverse transcription polymerase chain reaction( RT-PCR),and the protein expressions of HIF-1α and VEGF were measured by immunostaining in total 42 patients who were divided into invasive and non-invasive pituitary adenoma groups. The differences of HIF-1α and VEGF mRNA expression between different groups were compared by Pearson's correlation coefficient and the protein expression by Spearman's correlation coefficient. Results The protein expressions of HIF-1α and VEGF in invasive pituitary adenoma group were significant higher than those in non-nvasive pituitary adenoma group( χ2=9. 147,P 0. 05 and χ2= 5. 401,all P 〈0. 05). There were significant difference of VEGF mRNA expression between invasive and non-invasive pituitary adenoma groups( P〈 0. 05). The correlated analyses of mRNA and protein expression showed there was a negative relation between HIF-1α and VEGF in non-invasive pituitary adenoma groups( P 〈0. 05),and a positive relation in invasive pituitary adenoma groups( r = 0. 890,P 0. 05; r = 0. 436,P〈 0. 05). Conclusion The overexpressions of HIF-1α and VEGF protein and mRNA may play important role in invasiveness of pituitary adenomas.
