摘要 目的:探讨胃癌组织中缺氧诱导因子(HIF)-1α、Bax、Bcl-2蛋白的表达及其与肿瘤浸润、转移的关系。方法:应用免疫组织化学(PV9000)染色法检测100例胃癌组织中HIF-1α、Bax、Bcl-2蛋白的表达。结果:胃癌组织中HIF-1α、Bax、Bcl-2蛋白阳性表达率分别为56%(56/100)、40%(40/100)、68%(68/100),与阴性对照组的5%(1/20)、90%(18/20)、20%(4/20)相比,P均<0.05。Bax、Bcl-2蛋白在56例HIF-1α阳性表达的胃癌组织中的阳性表达率分别为17.9%(10/56)、89.3%(50/56),与在44例HIF-1α阴性表达的胃癌组织的阳性表达率68.2%(30/44)、40.9%(18/44)相比,P均<0.05。HIF-1α、Bax蛋白在胃癌组织中的表达呈负相关(r=-0.510,P<0.05),而HIF-1α、Bcl-2蛋白在胃癌组织中的表达呈正相关(r=0.515,P<0.05)。HIF-1α蛋白表达与胃癌组织浸润范围及有无淋巴结转移有关(P均<0.05),与分化程度无关(P>0.05)。结论:HIF-1α阳性表达胃癌组织中Bax低表达,Bcl-2高表达,提示HIF-1α可能通过减少促凋亡蛋白Bax的表达,增加抗凋亡蛋白Bcl-2的表达,参与胃癌的发生发展过程。HIF-1α蛋白表达与胃癌浸润转移密切相关。 Objective: To indentify the expression of HIF-1α and Bax,Bcl-2 proteins, and investigate the relationship between them and the invasion and metastasis of gastric carcinoma. Methods: The expres- sion of HIF-1α, Bax, Bcl-2 proteins were detected in 100 cases of gastric carcinoma by immunohistoehemistry (PV9000) staining. Results: The positive rates of HIF-1α, Bax, Bcl-2 proteins in gastric carcinoma tissues were 56% (56/100),40% (40/100),68% (68/100), compared with control groups 5% (1/20),90% ( 18/20 ), 20% (4/20), P〈0.05.Bax, Bc1-2 expression positive rates were 17.9% ( 10/56 ), 89.3 % (50/56) in 56 cases gastric carcinoma tissues of HIF-1α positive expression, compared with 68.2% (30/44),40.9% (18/44) in 44 cases gastric carcinoma tissues of HIF-1α negative expression,P〈0.05.The expression of HIF-1α and Bax protein in gastric carcinoma tissue had negative Correlation (r =-0.510, P 〈0.05),but the expression of HIF-1α and Bcl-2 protein in gastric carcinoma tissue had positive correlation (r=0.515, P〈 0.05).The expression of HIF-1α protein was correlated with infiltration range and lymph node metastasis (P〈0.05),but not related with cell differentiation grade (P〉0.05). Conclusion: HIF-1α may participate in the incidence and development of gastric carcinoma by reducing the expression of pro-apoptotic protein Bax and increasing anti-apoptotic protein Bcl-2 expression. The expression of HIF-1α protein is closely related with invasion and metastasis of gastric carcinoma.
