摘要
目的探讨坎离颗粒改善舒张性心力衰竭大鼠左室舒张功能的可能作用机制。方法 56只Wistar大鼠随机分为假手术组、模型组、中药组和缬沙坦组,每组14只。除假手术组外其余各组采用腹主动脉缩窄法制成心脏压力负荷超载心力衰竭模型。术后1周中药组给予坎离颗粒溶液6.75 g/(kg·d)灌胃,缬沙坦组给予缬沙坦混悬液7.2μg/(kg·d)灌胃,假手术组、模型组给予等量双蒸水灌胃,每日1次,共干预32周。观察各组大鼠心肌肌质网形态学变化、钙摄取功能、钙ATP酶2a(SERCA2a)活性及其蛋白表达。结果与假手术组比较,模型组大鼠心肌肌质网钙摄取速率显著降低,心肌组织SERCA2a活性降低,SERCA2a蛋白表达增加(P<0.05或P<0.01);心肌肌质网明显减少,线粒体空化严重。与模型组比较,中药组心肌肌质网钙摄取速率提高,SERCA2a活性、蛋白表达明显增高(P<0.05或P<0.01);心肌肌质网、线粒体结构完好。结论坎离颗粒可能通过增加心肌组织SERCA2a蛋白和活性、促进心肌肌质网钙摄取速率,从而改善舒张性心力衰竭大鼠左心室舒张功能。
Objective To explore the possible mechanism of Kanli Granule in improving the left ventricular diastolic function in rats with diastolic heart failure. Methods Fifty-six Wistar rats were randomly divided into sham operation group,model group,Chinese medicine group and valsartan group,with 14 rats in each group. Except for the sham operation group,the rest groups were treated with abdominal aortic constriction to make cardiac pressure overload induced heart failure model. One week after operation,Chinese medicine group were gavaged with Kanli Granule solution 6. 75 g /( kg·d),the valsartan group were gavaged with valsartan suspension 7. 2 μg /( kg·d),the sham operation group and model group were gavaged with double distilled water,1 time a day,a total intervention of 32 weeks. The changes of morphology,calcium uptake,the activity of SERCA2 a and protein expression in myocardium sarcoplasmic reticulum of all groups rats were observed. Results Compared with sham operation group,the calcium uptake rate of myocardial sarcoplasmic reticulum in model group was significantly decreased,SERCA2 a activity was decreased and SERCA2 a protein expression was increased in cardiac muscle tissue( P〈0. 05 or P〈0. 01); Myocardial muscle mass was significantly decreased,and the mitochondria were severely cavitation. Compared with the model group,the calcium uptake rate increased,SERCA2 a activity and protein expression were significantly increased in the Chinese medicine group( P〈0. 05 or P〈0. 01); Cardiac muscle sarcoplasmic reticulum and mitochondria keep good structure. Conclusion Kanli Granule can improve the left ventricular diastolic function in rats with diastolic heart failure probably by increasing the SERCA2 a protein and activity of the myocardium and promoting the calcium uptake rate of the myocardium sarcoplasmic reticulum.
