摘要
采用正交试验优化水提法提取海南苦丁茶中茶多糖,并研究了苦丁茶多糖在体外对人血红细胞自氧化和H2O2所致红细胞氧化溶血保护作用。结果表明:影响苦丁茶多糖提取率的因素主次关系为浸提温度>浸提时间>乙醇浓度>料液比。正交试验最佳条件为:浸提温度为85℃,乙醇浓度为75%,浸提时间为1.5h,料液比为1∶15,此条件下的多糖提取率为3.53%。海南苦丁茶多糖在体外能够明显抑制人血红细胞自氧化溶血作用,随着茶多糖浓度的增加抑制红细胞的自氧化溶血作用也逐渐增强,茶多糖浓度为200μg/m L-400μg/m L时,溶血度下降趋势平缓,当浓度大于400μg/m L时溶血度迅速下降,抗氧化活性迅速上升,当茶多糖浓度达到600μg/m L时与200μg/m L的Vc保护作用相近;茶多糖对H2O2所致红细胞的溶血度也具有保护作用,随着茶多糖浓度的增加,抑制H2O2所致红细胞的氧化溶血作用也逐渐增强,茶多糖的保护作用均比相同浓度下的Vc溶液弱。
Polysaccharides is extracted from Hainan Kuding tea by using the water extraction method with the technology optimized by designing the orthogonal tests. The protective effects of Kuding tea on hemolysis of red blood cells and H2O2 induced hemolysis are further studied. The result shows that the order of the elements affecting the extraction rate is temperature〉 time 〉ethanol concentration〉 ratio of material to solvent. The perfect condition for the extraction is Temperature: 85℃,Ethanol Eoncentration: 75%,Time: 1. 5h,and Ratio of Material to Solvent: 1∶ 1. 5. Under this condition the extraction ratio of Polysaccharides is 3. 53%. Polysaccharides extracted from Hainan Kuding tea has an obvious inhibiting effect on the hemolysis of red blood cells. The inhibiting effect increases with the increase of the density of Polysaccharides. The hemolysis effect declines slowly and steadily when the range of density of Polysaccharides is within 200μg / mL ~ 400μg / mL. But when the density is greater than 400 g / mL,the hemolysis effect declines rapidly and its antioxidant activity increases rapidly.The Vc antioxidant activities possess similar effects when density of Polysaccharides reach 600μg / mL and200μg / m. Polysaccharides extracted from Hainan Kuding tea has a protective effect on the hemolysis of red blood cells induced by H2O2. The inhibiting effect of the hemolysis of red blood cells also increases with the increase of the density of Polysaccharides. With the same densities,the antioxidant activity of Polysaccharides is weaker than Vc solution.
