雌二醇对香猪睾丸中SF-1和3β-HSD及NOS表达的影响

摘要 为探明外源雌二醇(E2)对香猪睾丸中类固醇生成因子-1(SF-1)、3β-羟类固醇脱氢酶(3β-HSD)和一氧化氮合酶(NOS)表达的影响,以便进一步探讨雌激素影响睾丸生殖功能的作用机制,将12头1日龄雄性贵州香猪随机平分为对照组(空白)、试验Ⅰ组、试验Ⅱ组和试验Ⅲ组,试验Ⅰ~Ⅲ组从2日龄连续9d经腹腔每头每天分别注射E20.1mL、0.5mL和2.5mL,26日龄时手术取出左侧睾丸,采用酶联免疫吸附法测定SF-1、3β-HSD及NOS的含量。结果表明:与对照组相比,试验Ⅰ~Ⅲ组睾丸中SF-1、3β-HSD和NOS的表达量显著升高(P〈0.05)。其中,SF-1的表达量随E2用量增加显著升高(P〈0.05);3β-HSD的表达量在E2用量为0.5mL/(d·头)时最高,在2.5mL/(d·头)时表达量又下降(P〈0.05);NOS的表达量各试验组间差异不显著(P〉0.05)。E2能够促进新生香猪睾丸中SF-1、3β-HSD及NOS的表达。 To investigate the effects of E2 on the expression of steroidogenic factor 1(SF-1),3β-hydroxysteroid dehydrogenase(3β-HSD)and nitric oxide synthase(NOS)in testis of Xiang pig and to explore mechanism of E2 affecting testis reproduction.Twelve male Xiang pigs,1day old,were randomly divided into control group(blank group),experimental group Ⅰ,experimental group Ⅱ and experimental group Ⅲ.Each pig of experimental group Ⅰ,Ⅱ,and Ⅲ was intraperitoneally injected different doses,0.1mL,0.5mL,and 2.5mL,of E2 for nine days respectively from 2days old.15 dafter the last injection(26days old),the left testis of each animal was removed by surgical operation.The content of SF-1,3β-HSD and NOS in testis was measured by Enzyme-linked immunosorbet assay(ELISA).Results:The expression level of SF-1,3β-HSD and NOS increased significantly(P 〈0.05)in experimental groups,compared with the control group.While the expression level of SF-1significantly increased(P〈0.05)with increasing E2 doses.The highest expression level of 3β-HSD was measured at 0.5 mL dose of E2(P〈0.05),but the expression level decreased(P 〈0.05)at 2.5 mL dose.The difference of the expression level of NOS was not significant between experimental groups(P〉0.05).E2 could increase the expression of SF-1,3β-HSD and NOS in testis of neonatal Xiang pigs.
