前列安丸对慢性非细菌性前列腺炎大鼠前列腺组织IL-1β、IL-10及TNF-α的影响 被引量:11

Effect of Qianliean Pill on IL-1β,IL-10,and TNF-α in Prostate Tissues of Chronic Nonbacterial Prostatitis Rats

摘要 目的观察前列安丸对慢性非细菌性前列腺炎(chronic nonbacterial prostatitis,CNP)大鼠模型炎性IL-1β、IL-10及TNF-α的影响,探讨其治疗CNP的作用机制。方法采用去势并注射苯甲酸雌二醇方法造成CNP大鼠模型,随机分成5组,模型组、阳性药普乐安片组及前列安丸高、中、低剂量组,每组10只,并以10只正常动物作为正常对照组。自去势第8日起,普乐安片组给予普乐安片10.80 g/kg灌胃,前列安丸高、中、低剂量组分别给予前列安丸11.00、5.50及2.75 g/kg灌胃,模型组及正常对照组给予2 mL/100 g体重蒸馏水灌胃,均每日1次至第30日。给药30日后处死大鼠,取出前列腺组织。计算前列腺指数,在光镜下观察大鼠前列腺病理改变,同时采用酶联免疫吸附法检测大鼠前列腺组织中白细胞介素-1β(IL-1β)、白细胞介素-10(IL-10)及肿瘤坏死因子α(TNF-α)水平。结果与正常对照组比较,模型组大鼠前列腺指数明显降低,IL-1β、IL-10及TNF-α水平显著升高(P<0.01);与模型组比较,前列安丸高、中剂量组和普乐安片组前列腺指数均明显降低(P<0.01);前列安丸各剂量组及普乐安片组IL-1β、IL-10及TNF-α水平明显降低(P<0.01)。与普乐安片组比较,前列安丸高剂量组TNF-α水平降低更明显(P<0.05)。与正常对照组比较,模型组大鼠前列腺发生明显炎性反应;与模型组比较,前列安丸低、中、高剂量组以及普乐安片组大鼠前列腺组织炎性反应较轻,病变程度有不同程度改善。结论前列安丸治疗CNP可能是通过调节前列腺局部免疫状态,减轻前列腺的炎症反应和降低前列腺组织中IL-β、TNF-α、IL-10水平来实现的。 Objective To observe the effect of Qianliean Pill(QP) on inflammatory factors such as IL-1β,IL-10,and tumor necrosis factor a(TNF-α) in chronic nonbacterial prostatitis(CNP) model rats,and to explore its therapeutic mechanism.Methods CNP rat model was established by castration and estradiol benzoate injection.Totally 50 rats were randomly divided into 5 groups,i.e.,the model group,the positive medicine group,the high dose QP group,the medium dose QP group,and the low dose QP group,10 in each group.Besides,10 normal rats were recruited as a normal control group.Since the 8th day of castration,Pulean Tablet(PT) at 10.80 g/kg was administered to rats in the positive medicine group by gastrogavage.QP at 11.00,5.50,and 2.75 g/kg was administered to rats in high,medium,and low dose QP groups by gastrogavage.Distilled water at 2 mL/100 g was administered to rats in the model group and the normal control group by gastrogavage,once daily for 30 successive days.After30 days of medication all rats were sacrificed and their prostate tissues were extracted.The prostatic index was calculated.Pathological changes of rat prostate were observed under light microscope.Meanwhile,levels of IL-1β,IL-10,and TNF-α were detected using enzyme linked immunosorbent assay.Resuits Compared with the normal control group,the prostate index obviously decreased,levels of IL-1β,TNF-α,and IL-10 in the prostate tissue significantly increased in the model group(P〈0.01).Compared with the model group,the prostate index obviously decreased in high and medium dose QP groups,and the positive medicine group(P〈0.01);levels of IL-1 β,TNF-α,and IL-10 obviously decreased in each QP group and the positive medicine group(P〈0.01).Compared with the positive medicine group,the TNF-αlevel decreased more obviously in the high dose QP group(P〈0.05).Compared with the normal control group,inflammatory reactions occurred obviously in rats' prostate of the model group.Compared with the model group,inflammatory reactions
