摘要
目的分析慢性非细菌性前列腺炎(CAP)大鼠的γ-干扰素(IFN-γ)、肿瘤坏死因子α(TNF-α)和白细胞介素10(IL-10)表达。方法选取广西医科大学动物实验中心提供的10只12月龄健康雄性SD大鼠,随机将其分为CAP模型组(n=5)和正常对照组(n=5)两组。建立动物模型并对其进行实验处理,运用免疫组化链酶卵白素-过氧化酶法检测前列腺组织IFN-γ、TNF-α蛋白表达,用双抗体夹心ELISA法对前列腺中IL-10表达进行定量分析,然后对两组大鼠前列腺病理形态学改变、IFN-γ、TNF-α和IL-10表达进行统计分析。结果正常对照组大鼠具有完整的前列腺组织结构,有少量粉红色正常分泌物存在于腺腔中,CAP模型组大鼠病理呈明显炎症表现。CAP模型组大鼠前列腺IFN-γ、TNF-α和IL-10表达[(0.327±0.034)pg/ml、(32.152±4.143)pg/ml、(330.140±31.480)pg/ml]均显著高于正常对照组[(0.018±0.002)pg/ml、(0.371±0.040)pg/ml、(0.015±0.001)pg/ml](P<0.05)。结论慢性非细菌性前列腺炎大鼠的IFN-γ、TNF-α和IL-10表达提升,在前列腺炎的发生发展过程中有这三种炎性因子参与,并对前列腺炎的转归造成直接影响,值得临床充分重视。
Objective To investigate the expressions of IFN-γ,TNF-α and IL-10 of rats with chronic nonbacterial prostatitis. Methods 10 twelve-month old healthy male SD rats provided by Guangxi Medical University animal experimental center were selected. They were randomly divided into CAP model group( n=5) and normal control group( n=5) two groups,the animal model was established and the experimental treatment was did,the prostate tissue IFN-γ,TNF-αprotein were detected by immunohistochemical strept avidin peroxidase method,the prostate IL-10 expression was quantitatively analyzed by double antibody sandwich ELISA method,then the prostate pathological changes,expressions of IFN-γ,TNF-αand IL-10 of the two groups were statistically analyzed. Results The normal control group rats had intact prostate tissue structure,and a few normal pink secretions were present in the glandular cavity. CAP model rats showed obvious inflammatory pathology.The expressions of IFN-γ,TNF-α and IL-10 in prostate of the CAP model group( 0. 327 ± 0. 034) pg / ml,( 32. 152 ± 4. 143) pg / ml,( 330. 140 ± 31. 480) pg/ml were significantly higher than those of the normal control group( 0. 018 ± 0. 002) pg/ml,( 0. 371 ± 0. 040) pg/ml,( 0. 015 ± 0. 001) pg/ml( P〈0.05). Conclusion The expressions of IFN-γ,TNF-α and IL-10 of rats with chronic nonbacterial prostatitis are increased,these three kinds of inflammatory factors are involved in the occurrence and development of prostatitis,and have direct and profound impacts on the outcomes of prostatitis. Therefore,the detection of these indexes is worthy of the clinical attention.
