非小细胞肺癌易感性与环氧合酶-2基因多态性的关系

Research on the relationship between cyclooxygenase-2 polymorphisms and non-small cell lung cancer susceptibility

摘要 目的 探讨环氧合酶-2(COX-2)基因多态性与非小细胞肺癌(NSCLC)易感性的关系.方法 我们收集了42例非小细胞肺癌患者和50例健康体检者的空腹静脉血,使用聚合酶链反应-限制性片段长度多态性(PCR-RFLP)分析两组COX-2基因多态性.结果 发现COX-2 rs689466的基因型AA在患病组的分布频率显著高于健康对照组(P<0.05),AA基因型比GG基因型患NSCLC的风险高出4.67倍.COX-2 rs5275的基因型CC在这两组中的分布有差异(P<0.05),CC基因型比TT基因型患NSCLC的风险高出7.02倍.对性别、年龄、吸烟和饮酒因素修正后,COX-2 rs689466的基因型AA和COX-2 rs5275的基因型CC依旧会增高NSCLC的易感性.对两个单核苷酸多态性(SNP)等位基因的连锁不平衡和单体型分析之后发现,患病组中A-C单体型的分布频率较对照组高,且两组比较差异有统计学意义(P<0.05).对性别、年龄、吸烟和饮酒因素修正后,两组A-C单体型的分布频率差异有统计学意义(P<0.05).结论 COX-2 rs689466和rs5275基因多态性与NSCLC的易感性相关,且A-C是NSCLC的易感单体型. Objective To explore the association between cyclooxygenase-2 (COX-2) polymorphisms and non-small cell lung cancer (NSCLC) susceptibility.Methods We collected fasting peripheral venous blood from 42 cases with NSCLC and 50 healthy controls through physical examinations,and used polymerase chain reaction-restricted fragment length polymorphism (PCR-RFLP) to analyze COX-2 polymorphisms of two groups.Results The distribution frequency of genotype AA of COX-2 rs689466 in case group was significant higher than that in control group (P < 0.05).Carriers with AA genotype were 4.67 times at risk of NSCLC than those with GG genotype.The distribution of genotype CC of COX-2 rs5275 was different between two groups (P < 0.05),and carriers with genotype CC were at 7.02 times higher risk of NSCLC than those with genotype TT.After corrected by sex,gender,smoking and drinking factors,AA genotype of COX-2 rs689466 and CC genotype of COX-2 rs5275 still contributed to increased risk of NSCLC.After analyzed of linkage disequilibrium (LD) and haplotypes of alleles in two SNPs,the distribution frequency of A-C haplotype in case group was higher than that in control group,with significant difference between two groups (P < 0.05).After corrected by sex,gender,smoking and drinking factors,statistical difference was still found in the total distribution of A-C haplotype between two groups (P < 0.05).Conclusion COX-2 rs689466 and rs5275 polymorphisms may be related to NSCLC susceptibility.And A-C haplotype might be a susceptibility haplotype for NSCLC.
