摘要
目的探讨miR-29c-5p在胆囊癌组织中的表达情况及其临床意义。方法采用实时定量PCR技术(stem-loop real-time reverse transcription-polymerase chain reaction, qRT-PCR)检测40例胆囊癌组织与癌旁组织中miR.29c.5p的相对表达量;分析miR-29c.5p表达与胆囊癌患者临床病理因素及预后的关系。结果与癌旁组织相比,胆囊癌组织中miR-29c-Sp表达明显降低(Z=-2.521,P=0.008).其表达与胆囊癌患者的淋巴结转移有关(X2=15.140,P〈0.001),而与年龄、性别、肿瘤大小、分化程度、浸润深度及肿瘤TNM分期无关(均P〉0.05)。MiR-29c-5p低表达与高表达的患者中位生存率分别为7.2个月和20.2个月,1年及3年生存率分别为32.0%、16.0%和55.7%、33.4%,差异有统计学意义(P=0.040)。MiR-29c-5p表达量不是影响胆囊癌患者生存的独立风险因素(P=0.974)。结论MiR.29c-5p在胆囊癌组织中表达下调,并可能与胆囊癌的淋巴结转移有关;miR-29c-5p低表达提示预后不良。
Objective To investigate the expression of miR-29c-Sp in patients with gallbladder cancer (GBC) and to analyze the association of the expression with the clinicopathologic characteristics. Methods The expression levels of miR-29c-5p in 40 patients with GBC along with their matched adjacent tissues were detected by using stem-loop real-time reverse transcription-polymerase chain reaction (qRT- qPCR). The relations of miR-29c-Sp expression with the clinicopathologic factors and prognosis of GBC patients were analyzed. Results Compared with tumor-adjacent tissue, the miR-29c-5p expression in GBC was significantly downregulated ( Z = - 2. 521, P = 0. 008 ). The expression of miR-29c-5p was closely associated with lynphatic metastasis (X2 = 15. 140, P 〈 0. 001 ), but irrelevant to age, sex, tumor size, differentiation, T stage and TNM stage ( all P 〉0. 05 ). The average survival period of low and high expression of miR-29c-5p were 7.2 months and 20. 2 months. The one-year and three-year survival rate of patients with low expression of miR-29c-5p were significantly lower than those with high levels (32.0% , 16.0% vs 55.7%, 33.4% ) ( P =0. 040). Low miR-29c-5p expression is not risk factor affecting the survival of GBC patients (P = 0. 974 ). Conclusions The miR-29c-5p expression is downregulated in GBC tissue, which probably may be associated with the metastasis of GBC, and low miR-29c-Sp expression may predict an unfavorable outcome.
