摘要
观察红三叶草异黄酮对高糖高脂饮食诱导的脂肪肝大鼠脂质过氧化及肝功能的影响。雄性SD大鼠54只,饲养一周后,随机抽取12只大鼠为对照组,喂普通饲料。其余为造模组,脂肪乳灌胃,同时给予18%的蔗糖溶液。连续4周后,随机处死2只对照组大鼠和2只造模组大鼠,做肝脏病理切片,镜下见脂肪变,表明造模成功。然后将造模组大鼠按体重随机分为4组(每组10只):分别为模型组、红三叶草异黄酮低、中、高剂量组(75、150、300 mg/kg灌胃),共计12周。实验结束处死大鼠,收集血清和肝组织,检测血清及肝组织中总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白胆固醇(LDL-C)、高密度脂蛋白胆固醇(HDL-C)、游离脂肪酸(FFA)、超氧化物歧化酶活性(SOD)、丙二醛(MDA)、谷丙转氨酶(ALT)、谷草转氨酶(AST)的活性或含量。与模型组比较,红三叶草异黄酮中、高剂量组能明显降低血清及肝脏中TC、TG、LDL-C及FFA的含量,升高血清及肝脏中HDL-C的含量,血清及肝组织SOD酶活性升高,MDA含量降低(P﹤0.05);肝功能指标血清ALT及AST(145.88±34.28)的含量明显低于模型组(P﹤0.05)。一定剂量的红三叶草异黄酮具有调节脂质代谢及减少高糖高脂膳食导致的氧化损伤作用,肝细胞损伤减轻,肝功能改善,对非酒精性脂肪肝大鼠肝功能具有一定的保护作用。
Observation of Red Clover Isoflavones on liver function and inflammatory response of nonalcoholic fatty liver disease in rats. After one week feeding in laboratory , 12 out of 54 male SD rats were randomly chosen as the normal control group and fed with normal diet. The rest were made as modules , gavage feeding with fat emulsion and 18 % sucrose solution. Two rats from normal control group and two from module were killed randomly after 4 weeks. Liver biopsy showed the module formation was successful. Then we randomly divided module into 4 groups (n=10) according to weight:Nonalcoholic fatty liver diseases (NAFLD) model control group, high-dose, medium-dose and low-dose Red Clover Isoflavones treatment group (75, 150, and 300 mg/kg), respectively. The experiment period lasted 12 weeks. At the end of experiment , rats were killed to collect serum and liver tissues. Then we detected the activity and content of Aspartate aminotransferase (AST), alanine aminotransferase (ALT), total cholesterol (TC), triglyceride (TG), low density lipoprotein cholesterol (LDL-C), high density lipoprotein cholesterol (HDL-C), malondialdehyde (MDA), superoxide dismutase (SOD), alanine aminotransferase (ALT), aspartate aminotransferase (AST) and free fatty acids (FFA). Compared with model group, the middle-dose group and the high-dose group could significantly decrease TC, TG, LDL-C and FFA levels in serum and liver tissues and increase HDL-C levels. Compared with the model group , the middle-dose group and the high-dose group of red clover isoflavones could decrease MDA levels and improve SOD activity in serum, and the differences were statistically significant (p〈0.05). The levels of ALT and AST (145.88±34.28) in middle and high dose group were lower than control group (p〈0.05).
