摘要
研究了室温制备的龙须菜多糖GCp的分离纯化和生物学活性。采用超滤分级将GCp分为两部分GCp F1和GCp F2;采用离子凝胶层析方法,进一步分离了GCp F2,获得4个组分,发现其主要由中性多糖GCp F2-1组成,回收率24.32%;离子层析纯化后4个组分的多糖纯度提升显著,其中GCp F2-1多糖含量最高,达74.29%。刺激小鼠脾淋巴细胞增殖试验显示,GCp F2-1活性显著,呈现剂量依赖关系,与GCp和GCp F2相当,而其余3个组分多糖的活性均有所下降。同时,发现龙须菜多糖在体外不具有抑制肿瘤细胞L1210增殖作用。
The isolation and bioactivity of polysaccharide GCp,which was extracted from Gracilaria lemaneiformis at room temperature,was studied.GCp was isolated to GCpF1 and GCpF2 by ultrafiltration.Ion chromatography was applied to further isolation of GCpF2.The results showed that GCpF2 was composed of 4 parts,including neutral polysaccharide GCpF2-1 which was the mainly component.After ion chromatography,the recovery rate of GCpF2-1 was 24.32% ,and the polysaccharide content of 4 samples were significantly improved.The polysaccharide content of GCpF2-1 reached the highest level of 74.29% .The research indicated that GCp,GCpF2 and GCpF2-1 had the similar activity on the assay of inducing proliferation of mouse lymphocyte,but the activities of the other 3 samples were descendant.The study also showed that the polysaccharides form Gracilaria lemaneiformis didn't have the activity of inhibition of tumor cells L1210 in vitro.
