Optimization of Extraction Technology Based on Central Composite Design-Response Surface Methodology for Paeoniae Radix Rubra

摘要 目的优选赤芍中芍药苷的提取工艺。方法采用星点设计-效应面法,以芍药苷的提取率为指标进行试验,考察乙醇浓度、乙醇体积、超声时间对提取工艺的影响,并进行二次多项式拟合,预测最优工艺参数。结果确定最佳提取工艺为:乙醇浓度76%,乙醇体积13倍,超声时间55min,提取2次,芍药苷提取率为3.6831%。结论星点设计-效应面法可用于赤芍提取工艺的优选,所建立的提取工艺稳定可靠。 OBJECTIVE To optimize the process of extracting paeoniflorin from paeoniae radix rubra. METHODS The influential factors were ethanol concentration,ethanol volume and ultrasonic time. The evaluating indicator was the extraction rate of paeoniflorin. The central composite design-response surface methodology was used to optimize the process and the prediction was carried out. RESULTS The optimum conditions of extraction were 76% ethanol,13 volume folds of solvent and 55 min for two extraction. CONCLUSION This method is suitable for optimizing the extraction of paeoniflorin in paeoniae radix rubra,and the extraction process is stable and reliable.
