摘要
【目的】探讨蓝莓皮渣综合利用的基础因子。【方法】以花色苷含量为试验指标,考察了乙醇体积分数、料液比、p H值、提取温度及提取时间等因素对提取效果的影响。在单因素试验的基础上,利用响应面分析法优化提取工艺条件,得到适宜的工艺条件。通过测定蓝莓皮渣花色苷的总抗氧化能力、清除DPPH·和OH·能力,评价其抗氧化活性。【结果】研究结果表明,提取时液料比、提取时间、提取温度、p H、乙醇体积分数对提取量影响较大,通过响应面分析法优化了提取工艺,其适宜的提取工艺条件为:乙醇体积分数60%、V(液,m L)、m(料,g)比20∶1、提取时间1.5 h、p H 2.0、提取温度35℃,在该工艺条件下,蓝莓渣花色苷的提取量为16.197 mg·g-1;蓝莓皮渣花色苷具有一定抗氧化能力,在实验范围内,DPPH·和OH·的抑制率均随质量浓度的升高而增加,清除DPPH·和OH·的IC50分别为241.88和1 238.15μg·m L-1。【结论】优化了蓝莓皮渣花色苷提取的工艺条件,且蓝莓皮渣花色苷具有一定的抗氧化活性。蓝莓皮渣花色苷的提取及抗氧化活性分析为其综合利用奠定了基础。
【Objective】The objective of the study is to develop the basis for comprehensive utilization ofanthocyanin from blueberry pomace.【Methods】The influences of ethanol concentration,solvent-materialratio,time,p H and temperature on the anthocyanins content were investigated in single-factor experi-ments. The process parameters were optimized by response surface analysis method. In addition,the anti-oxidant activity of anthocyanin was evaluated based on total antioxidant power,DPPH and OH radical-scavenging assay.【Results】The results showed that ethanol concentration,solvent-material ratio,time,p H and temperature had great impact on content of the anthocyanins,the optimum extraction conditionwere ethanol concentration 60%,25∶1 of solvent-material ratio,1.5 h of time,p H 2.0 and 35 ℃ of tem-perature by response surface analysis method. The content of the anthocyanin was 16.197 mg·g- 1by themethod. The anthocyanin from blueberry pomace displayed dose-dependent reducing power and abilitiesto scavenge DPPH and hydroxyl free radicals,and IC50 values of DPPH·and OH· were 241.88 mg·L- 1and 1 238.15 mg·L- 1.【Conclusion】The extraction condition of anthocyanin from blueberry pomace wasoptimized and anthocyanins from blueberry pomace showed good antioxidant activities.
