Identification and Determination of Sweeteners, Preservatives and Pigments in Beverage by High Performance Liquid Chromatography with Diode Array Detector

摘要 旨在利用高效液相色谱配备二极管阵列检测器同时鉴别并测定饮料中甜味剂、防腐剂和色素。方法采用Waters Sun Fire C18柱(4.6 mm×250 mm,5μm)进行分离,以甲醇与乙酸铵(0.02 mol/L)体系进行梯度洗脱,二极管阵列检测器检测样品,利用紫外光谱图进行定性分析,外标法进行定量分析。结果表明,质量浓度在1~50μg/mL时,各组分校准曲线的相关系数均大于0.999;检测限为:苯甲酸0.17μg/g、山梨酸0.04μg/g、脱氢乙酸0.14μg/g、糖精钠0.57μg/g、柠檬黄0.34μg/g、苋菜红0.04μg/g、胭脂红0.05μg/g、日落黄0.03μg/g、诱惑红0.04μg/g;样品加标回收率范围为90%~110%;相对标准偏差(RSD)≤8%。表明该方法具有快速、准确、操作简单等优点。 The purpose is to identification and determination of sweeteners, preservatives and pigments in beverage by high performance liquid chromatography with diode array detector. The analysis was performed on a Waters Sun Fire C18 column (4.6 mm×250 mm, 5 μm), using methanol and (0.02 mol/L) acetic acid ammonittm system for gradient elution by diode array detector to test sample, ultraviolet spectrum for qualitative analysis, extema/standard method for quantitative analysis. The results showed that the concentration between 1-50 μg/mL; the correlation coefficient of standard curve were greater than 0.999; Detection limit was that benzoic acid 0.17 μg/g, sorbic acid 0.04 μg/g, dehydroacetic acid 0.14 μg/g, saccharin sodium 0.57 μg/g, citric yellow 0.34 μg/ g, amaranth 0.04μg/g, carmine 0.05 μg/g, sunset yellow 0.03 μg/g, and the temptation to red 0.04 μg/g, respectively. Recoveries for samples was 90%-110%; RSD ≤ 8%. The method is rapid, accurate, and operates simply.
