摘要
目的建立豚鼠形觉剥夺性近视眼模型,研究视网膜转化生长因子-β_2(TGF-β_2)mRNA、胰岛素样生长因子-Ⅰ(IGF-Ⅰ)mRNA在形觉剥夺性近视眼的变化及作用。方法出生3d的实验用花色豚鼠45只,半透明眼罩遮盖左眼,右眼为对照眼。遮盖4周实验结束时,RT-PCR检测后极部视网膜中TGF-β_2mRNA、IGF-ⅠmRNA的表达水平。结果遮盖4周后遮盖眼形成明显的近视,TGF-β_2mRNA在遮盖眼表达量为(0.437±0.032),对照眼为(0.671±0.048),遮盖眼表达量较对照眼下降,差异有高度统计学意义(t=11.653,P<0.01);IGF-ⅠmRNA在遮盖眼表达量为(1.817±0.144),对照眼为(1.536±0.113),遮盖眼表达量较对照眼升高,差异有高度统计学意义(t=7.194,P<0.01)。结论 TGF-β_2、IGF-Ⅰ参与豚鼠形觉剥夺性近视形成。
Objective To explore the effects of retinal transforming growth factor-beta 2 (TGF-β2) and insulin-like growth factor-1 (IGF-Ⅰ) in form-deprivation myopia guinea pig model. Methods 45 three-day-old guinea pigs were selectedall left eyes were coverred with transplant goggles, and the right eyes were control eyes with no goggles. Refrac-tive status was measured by strip light retinascopy at before experiment and after 4 weeks. Total RNA in the posterior retina was extracted at the end of 4 weeks, the expression levels of TGF-β2 mRNA and IGF-ⅠmRNA were measured by PCR. Results Form-deprivation eyes were significantly more myopia than control eyes,. the expression level of TGF-β2 mRNA in form-deprivation eyes was (0.437±0.032), that in control eyes was (0.671±0.048), two groups were compared, the difference was statistically significant (t =11.653, P〈0.01). The expression level of IGF-Ⅰ mRNA in form-deprivation eyes was (1.817±0.144), that in control eyes was (1.536±0.113), two groups were compared, the dif-ference was statistically significant (t=7.194, P〈0.01). Conclusion TGF-β2 and IGF-Ⅰ participate in the develop-ment of form deprived myopia in guinea pig.
