番石榴组织培养的初步研究

摘要 以番石榴无菌茎段为材料,研究了生长调节剂及MS强度对番石榴无菌茎段芽萌发和茎段生根的影响。结果表明,以MS为基本培养基时,最有利于番石榴无菌苗生长的NAA浓度为0.4 mg·L-1,叶片数(7.59片)、节数(3.95节)、苗高(5.10 cm)和节间长度(1.41 cm)最大;6-BA浓度为5.0 mg·L-1时,茎段具有最高的芽诱导率(100%)和诱较高的芽诱导数(2.44个);1/2 MS+NAA 0.05 mg·L-1最有利于番石榴茎段的生根,生根率100%,根数2.30条,根长4.64 cm。在番石榴组织培养中,NAA能够促进番石榴茎段的生长,但会抑制芽的诱导。 In our paper, sterile stem segments of guava were selected as explants to study effects of plant growth regulators and MS culture medium strengths on bud induction and rooting in vitro. The resuhsindicated that, when using MS as the basic medium, best concentration of NAA for seedling growth in vitro was 0.4 mg ·L-l, with the highest value of leaf number (9.59 leaves) , node number (3.95 nodes) , seedling height (5.10 cm) and node length (1.41 cm) among media studied; when the concentration of 6-BA was 5.0 mg·L-1, bud inducing rate was 100% and number of buds induced was 2.44. Medium of 1/2 MS + NAA 0.05 mg·L-1 was best for rooting of guava seedling in vitro, with a rooting percentage of 100%, root number of 2.30 and root length of 4.64 cm. For the tissue culture of guava, NAA could enhance growth of stem segment, but inhibited bud induction.
