摘要
柚子是1种营养价值极高的水果,其肉与皮,均富含胡萝卜素、B族维生素、维生素C、矿物质、糖类、蛋白质、有机酸等,对保持人体健康,防病治病具有重要的作用。维生索C又称L-抗坏血酸(ascorbic acid),是含有6碳的不饱和多羟化合物,在医药及营养保健方面的用途十分广泛。维生素C的分析方法有多种,但这些方法往往操作复杂费时,在样品处理过程中很容易使维生素C损失,且灵敏度和精确度不高;而高效液相色谱法(HPLC)具有较高的准确度、精密度和灵敏度,简便快速,更加通用于维生素C的测定。本研究采用超声波提取法从柚子果肉中提取维生素C,通过高效液相色谱法,探究柚子果肉中维生素C的最佳分析条件。通过大量的实验研究,表明以甲醇:纯水=1:9的混合液为流动相,流动相的速度为0.6ml/min,检测波长为267 nm,柱温在25℃为柚子果肉中维生素C的最佳HPLC分析条件。在此条件下分析柚子果肉中维生素C的含量、稳定性、回收率,所建立的标准曲线具有良好的线性关系,其相关系数R^2=0.9994;其平均回收率为99.52%,重复性相对标准偏差小于0 63%;测得柚子果肉中维生素C的含最为120.65 mg/100 g。所建立的方法简单、快速、准确度高,适宜柚子果肉中维生素C含量的分析研究。
The pomelo pulp's nutrition is comprehensive and abundant,its pulp and peel contains abundant of carotene,B group vitamins,vitamin C, minerals,carbohydrate,protein and organic acids,etc.It plays an important role in maintaining human health,disease prevention and control.Vitamin C,also known as L-ascorbic acid(ascorbic acid),that contains 6 carbons unsaturated hydroxyl compounds,pharmaceutical and nutritional care in the use of wide-ranging.There are some methods for analyzing vitamin C,but these methods are often complex and time-consuming operation,the sample processing is very easy for the loss of vitamin C,and the sensitivity and accuracy is not high.However,the high performance liquid chromatography(HPLC) with high accuracy,precision and sensitivity,it is simple,rapid and more suitable for the determination of vitamin C.In this study,vitamin C was extracted from pomelo pulp by ultrasonic extraction method,the optimum HPLC conditions of vitamin C in pomelo pulp were explored.In this way,the content,the repeatability and the retums-ratio of vitamin C in pomelo pulp were analyzed under the optimum condition of that the mixture of methanol mixed with pure water by 1:9 by volume was taken as flowing,and its speed was 0.6 ml/min,and the ultraviolet examination wave length was 267 nm with the column temperature under 25℃.The good linear relations were gotten through the linear return with its correlation coefficient being R^2=0.9994.Its average returns-ratio was 99.52%.The repeatability's relative standard deviation was smaller than 0.63%.The content of vitamin C in pomelo pulp was 120.65 mg/100 g.The method established was simple,fast and accurate,which is suitable to analyze the content of vitamin C in pomelo pulp.
