摘要
目的:建立测定僵蚕中草酸铵含量的方法。方法:采用微流控芯片非接触电导法,探讨缓冲液种类和浓度、添加剂种类和浓度、进样时间和分离电压等因素对分离检测的影响。结果:优选缓冲体系为3 mmol/L三羟甲基氨基甲烷+3 mmol/L柠檬酸(p H=3.0)、不加添加剂、进样时间为10 s、分离电压为2.0 kV。草酸铵质量浓度在10~150μg/ml范围内与峰面积呈良好线性关系(r=0.999 8);检测限为3.0μg/ml;精密度、稳定性、重复性试验的RSD均〈2.0%;平均加样回收率为99.1%,RSD=1.4%(n=9)。结论:该方法简便快速、重复性好,为僵蚕的质量控制提供了一种新方法。
OBJECTIVE:To establish the method for the content determination of ammonium oxalate in bombyx batryticatus.METHODS:Microfluidic chip with contactless conductivity were conducted to detect the effects of types and concentration of buffer solution and additives,volume time and separation voltage on separation detection.RESULTS:The butter system was optimized as3 mmol/L Tris+3 mmol/L citric acid(p H=3.0)with no additives,volume time of 10 s and the separation voltage of 2.0 kV.There was a good linear relationship between the mass concentration of ammonium oxalate and peak area in the range of 10-150 μg/ml(r=0.999 8)with the detection limit of 3.0 μg/ml;the RSDs of precision,stability and reproducibility tests were less than 2.0% and the average recovery was 99.1%(RSD=1.4%,n=9).CONCLUSIONS:The method is simple,rapid and well reproducible and provides a new method for the quality control of bombyx batryticatus.
