三七总皂苷对局灶性脑缺血大鼠不同恢复时点神经重塑和突触重建的影响 被引量:14

Influence of Panax Notoginseng Saponins on neuronal plasticity and synaptic reorganization of rats with focal cerebral infarction in different recovery timing

摘要 目的:观察三七总皂苷(PNS)对局灶性脑缺血大鼠不同恢复时点Rho A、Rock2、Syp和PSD-95蛋白表达的影响,以探索PNS对神经重塑与突触重建的作用机制。方法:采用健康雄性SD大鼠,将其分为假手术组、手术组和Y27632组、PNS加Y27632组,对除假手术组外所有动物采用Longa法建立大脑中动脉阻塞MCAO模型,再将手术组动物随机分为模型组、PNS组、尼莫地平组,每组均为6只,利用HE染色和免疫组化方法观察术后14d和28d大鼠皮层梗死周围区Rho A、Rock2、Syp和PSD-95蛋白表达及梗死周围区形态学变化。结果:MCAO术后14d和28d,均可见模型组大鼠Rho A和Rock2蛋白的表达升高(P<0.01),Syp和PSD-95蛋白表达下降(P<0.01);各用药组中Rho A和Rock2蛋白的表达(除28d尼莫地平组的Rock2外)均比模型组减少(P<0.05,P<0.01),术后Syp和PSD-95蛋白(除14d尼莫地平组的Syp外)与模型组相比增加(P<0.05,P<0.01)。结论:PNS可抑制脑梗死后Rho A和Rock2蛋白表达并促进Syp和PSD-95蛋白表达,从而促进神经重塑和突触重建。 Objective: To observe the influence of Panax Notoginseng Saponins (PNS) on protein expression of RhoA, Rock2, Syp and PSD-95 of rats with focal cerebral infarction in different recovery timing, in order to investigate the mechanism of PNS on neuronal plasticity and synaptic reorganization. Methods: Healthy male SD rats were randomly divided into sham-olberated group, operated group, Y27632 group, PNS plus Y27632 group. Middle cerebral artery occlusion (MCAO) rat models were established by using Longa method of all the rats except sham-operated group. Rats in operated group were randomly divided into model group, PNS group and Nimodipine group with 6 rats in each group. HE staining and immunohistochemical method were used to observe the morphological changes and protein expression of RhoA, Rock2, Syp and PSD-95 around the cortex infarction of rats on the 14th day and 28th day after operation. Results: On the 14th day and 28th day after MCAO operation, the expression of RhoA and Rock2 protein of rats in model group was increased (P〈0.01), and the expression of Syp and PSD-95 protein was decreased (P〈0.01). Compared with model group, the expression of RhoA and Rock2 protein of rats in the treatment groups was decreased (P〈0.05, P〈0.01). Compared with model group, the expression of Syp and PSD-9 protein was increased after operation (P〈0.05, P〈0.01), in addition to the expression of Syp in Nimodiping group at 14th day. Conclusion: PNS could inhibit expression of RhoA and Rock2 protein and promote expression of Syp and PSD-95 protein after cerebral infarction, thereby, it could promote neuronal plasticity and synaptic reorganization.
