摘要
目的:观察脂肪干细胞(adipose-derived stem cell,ADSC)移植对局灶性脑缺血大鼠大脑皮质RhoA和Rock2表达的调节作用。方法:将大鼠随机分为假手术组(sham组)、模型组(model组)和ADSC组。体外分离培养ADSC,线栓法制作大鼠大脑中动脉栓塞模型。ADSC组于造模成功后经侧脑室注射ADSC悬液,模型组注射磷酸盐缓冲液。各组大鼠分别于7 d和14 d处死取材,采用RT-PCR法和Western blot法检测大脑皮质RhoA和Rho相关激酶2(Rho-associated protein kinases 2,Rock2)表达变化。结果:模型组和ADSC组各个时间点大脑皮质RhoA和Rock2表达水平较假手术组增多(P<0.05);ADSC组各个时间点大脑皮质RhoA和Rock2表达水平较模型组减少(P<0.05)。结论:ADSC移植可抑制脑缺血损伤后RhoA和Rock2的表达,从而促进神经功能重塑。
Objective : To observe the effect of adipose - derived stem cell(ADSC) transplanting on the expression of RhoA and Rock2 in rats with focal cerebral ischemia. Methods:Rats were randomly divided into sham operation group( sham group), middle cerebral artery occlusion group (model group) and ADSCs -treated group( ADSC group). ADSCs were harvested and purified and rat focal cerebral ischemia models were established by the suture method. One day after right middle cerebral artery occlusion(MCAO) ,ADSCs were injected into the lateral ventricle of ADSC group rats. When the rates were killed after 7 d and 14 d reperfusion, the expressions of RhoA and Rock2 were detected by RT - PCR and Western blot. Results: The expressions of RhoA and Rock2 in model group and ADSC group were significantly higher than those in sham group at each time point ( P 〈 0.05 ) ; the expressions of RhoA and Rock2 in ADSC group were lower than those in model group at each time point ( P 〈 0.05 ). Conclusion:ADSC transplanting can promote neuronal plasticity, partly by inhibiting the expressions of RhoA and Rock2.
