铁皮石斛根提取物对2型糖尿病模型小鼠的降糖作用研究 被引量:43

Hypoglycemic effect of extracts from Dendrobium officinale roots on type 2 diabetic model mice

摘要 目的:铁皮石斛的传统药用部位(茎)有"滋阴清热"的功效,与糖尿病的防治关系密切。目前大量的铁皮石斛根作为非药用部位被废弃,造成了资源的大量浪费。本实验采用高糖高脂加链脲佐菌素诱导小鼠模型,观察铁皮石斛根提取物对2型糖尿病模型小鼠的降糖作用,为铁皮石斛根的充分利用提供药理学依据。方法:采用高糖高脂饲养8 w加腹腔注射链脲佐菌素2次致2型糖尿病小鼠模型。造模10 w后,灌胃给予铁皮石斛根不同溶剂的提取物(EW、EA和EB),2次/日,单次给药剂量为5 g/kg,连续给药10 w。期间观察小鼠体征、体重、饮水和摄食;给药第6、8、10 w测定空腹血糖,给药第8 w测定口服糖耐量;给药第10 w测定糖化血红蛋白(Hb A1c)、血清胰岛素(INS)和胰岛素受体β(ISR-β),计算胰岛素敏感指数(ISI)。结果:⑴和模型组相比,3种提取物均能不同程度地改善模型小鼠的体征,减少饮水和摄食,其中提取物EB的改善效果较明显。⑵和模型组相比,提取物EB能明显降低模型小鼠的空腹血糖(P〈0.05),给药6、8、10 w,血糖值分别由模型组(19.7±4.7)mmol/L降至(15.1±4.6)mmol/L、由(19.5±4.7)mmol/L降至(15.1±4.5)mmol/L、由(19.3±5.9)mmol/L降至(13.3±5.5)mmol/L;糖耐量实验中,给药8 w,提取物EB能有效降低模型小鼠给予葡萄糖前后各时间点的血糖值和血糖-时间曲线下面积(P〈0.05~0.01),提取物EA能明显降低小鼠糖负荷后2 h的血糖值(P〈0.05)。⑶进一步研究发现提取物EB能显著降低模型小鼠的HbA 1c(P〈0.01),明显提高ISI(P〈0.05)。结论:铁皮石斛根提取物有改善2型糖尿病模型小鼠的作用,其中提取物EB降糖效果明显且平稳,并能有效改善小鼠的体征、生活质量和口服糖耐量,一定程度上为根部的合理利用和石斛资源的综合开发提供了新的思路。初步研究发现提取物EB的降 Objective: It has been shown that the effect of " nourishing yin and clearing heat" from traditional medicinal stem of Dendrobium officinale( D. officinale) were closely related with the prevention and treatment of diabetes relations. Now huge non-medicinal root were wasted,resulting in a large waste of resources. Therefore,the hypoglycemic effect of extract from D. officinale roots on type 2 diabetic model mice which fed with enrich-diet and streptozotocin was investigated,and to provide pharmacology basis for fully utilization of D. officinale roots.Methods: Type 2 diabetic model was established by feeding eight-week high sucrose fat diet and abdominal injecting of streptozotocin( STZ) twice. After 10 w of modeling,they were orally administrated with extract in different solvents( EW,EA and EB) of D. officinale roots of single dose of 5 g / kg,twice daily,lasted for 10 weeks. During the experiment,sign,weight,food and water intake were measured.Fasting blood glucose( FBG) was determined in the 6th,8th and 10 th weeks. Oral glucose tolerance test( OGTT) was measured in the 8th week. The indexes such as glycosylated hemoglobin( Hb A1c),insulin( INS) and insulin receptor-beta( ISR-β) were measured and the insulin sensitivity index( ISI) was calculated in the 10 th week. Results: ⑴Compared with model group,three extracts could improve the sign and reduce the water and food intake of model mice,and the extract EB was more obvious. ⑵Compared with model group,extract EB could obviously lower FBG of model mice( P 〈0. 05),and the value respectively were by model group( 19. 7 ± 4. 7) mmol / L down to( 15. 1 ± 4.6) mmol / L,by( 19. 5 ± 4. 7) mmol / L down to( 15. 1 ± 4. 5) mmol / L,by( 19. 3 ± 5. 9) mmol / L down to( 13. 3 ± 5. 5) mmol / L in the6 th,8th and 10 th weeks. During the OGTT,extract EB could effectively reduce the blood glucose levels of three time points and the area under curve( P 〈0. 05 ~ 0. 01) and extract EA could
