摘要
目的:探讨小檗碱与苦参总碱不同配比对溃疡性结肠炎大鼠模型的治疗作用。方法:采用2,4-二硝基氯苯复制大鼠溃疡性结肠炎模型,检测大鼠粪便常规、隐血试验、长度8cm病变结肠的粘连指数和重量,并进行结肠黏膜组织病理学检查,测定黏膜组织肿瘤坏死因子、环氧合酶-2的表达。结果:小檗碱180mg/kg、小檗碱120mg/kg+苦参总碱60mg/kg给药3天后能明显改善溃疡性结肠炎大鼠粪常规,隐血试验大部分呈阴性;给药7天后结肠腺体多数完整,改善结肠组织病理学检查;结肠黏膜肿瘤坏死因子及环氧合酶-2平均灰度较模型组有显著性差异。结论:小檗碱180mg/kg、小檗碱120mg/kg+苦参总碱60mg/kg对2,4-二硝基氯代苯致溃疡性结肠炎大鼠治疗效果显著。
Objective: To discuss the therapeutic effect of different compatibility proportion of berberine and total matrines on ulcerative colitis model rats. Methods: Ulcerative colitis model rats were induced with 2,4-nitro chlorinated benzene( DNCB). Detect rats' dung routine and conduct occult blood test,8cm length of the adhesion index and colon weight,pathological changes of colonic mucosa tissue,tumor necrosis factor and expression of cyclooxygenase-2 were observed significant differences with the model group,P 〈0. 01. Results: Berberine group( berberine,180 mg / kg) and composite group 4( berberine120 mg / kg + total matrines 60 mg / kg),of which most rats' occult blood test is negative,can significantly improve rats' dung routine for three days; most rats' colonic gland were complete,the colon histopathological examination improved obviously,and the expression of TNF-α and COX-2 reduced significantly. Conclusion: Berberine 180 mg / kg and berberine 120 mg /kg + total matrines 60 mg / kg has some therapeutic effect on UC rats and the effect is remarkable.
