离子色谱-脉冲安培法分析西红花多糖的单糖组成 被引量:4

摘要 采用水提醇沉法提取西红花球茎、柱头、花瓣、侧芽等部位的粗多糖,离心干燥得到的水溶性粗多糖在沸水浴条件下经硫酸水解后,通过离子色谱-脉冲安培法测定西红花不同部位多糖的单糖组成.该方法简单高效,灵敏度高,选择性好,不需要气相色谱和毛细管电泳所需的衍生化操作.在优化条件下,测得西红花多糖中含有岩藻糖、鼠李糖、阿拉伯糖、甘露糖、葡萄糖、半乳糖和果糖7种单糖.采用此方法,各单糖检测的线性范围在0.02~20mg·L-1,线性相关系数均大于0.999 2,加标回收率在87.2%~111.6%,相对标准偏差在1.26%~3.61%,检出限在3.70~11.35μg·L-1. The water-soluble crude polysaccharides were extracted from the safflower with hot water followed by precipitating with ethanol and drying. Then, the polysaccharides were hydrolyzed with sulfuric acid in the boiling water. The monosaccharide composition of polysaccharides from the safflower was analyzed by ion chromatogra- phy-pulsed amperometric detection. This method is simple, sensitive and selective without derivatization. Under the optimum conditions, our results show that the polysaccharide fraction is mainly composed of fucose, rhamnose, ara- binose, mannose, glucose, galactose and fructose. The linear ranges of this method for these monosaccharides are in the range of 0.02 to 20 mg L-1 with the correlation coefficients r≥0. 999 2, whereas the detection limits calculated at S/N=3 are in the range of 3.70 to 11.35 g L-1. The recoveries are in the range of 87.2% to 111.6% with the relative standard deviations are from 1.26 % to 3.61%.
