摘要
旨在建立一种能够同时测定牛肉水解产物中葡萄糖、果糖、阿拉伯糖、半乳糖、甘露糖、核糖的离子色谱检测方法。对样品的前处理与色谱条件进行优化,采用1%三氯乙酸溶液沉淀蛋白和4 mol/L硫酸溶液水解条件,选择CarboPacTM PA20离子色谱柱,用5 mmol/L氢氧化钠溶液为淋洗液进行洗脱,用脉冲安培检测器PAD进行测定分析。结果表明,6种糖在上述条件下能够完全分离,葡萄糖在0.25~2.50μg/mL、其他糖在0.025~0.25μg/mL区间内线性良好,相关系数r均大于0.998。方法精密度RSD在2.3%~5.5%,回收率在79.6%~116.5%之间。该方法操作简便、分离度好,能够准确、快速地测定牛肉水解产物中糖的含量。
The aim of this study is to establish an ion chromatography method for simultaneous determination of glucose,fructose,arabinose,galactose,mannose and ribose in beef hydrolysates.The pretreatment and chromatography conditions were optimized.Finally,the protein was precipitated by 1%trichloroacetic acid and polysaccharide was hydrolyzed by 4 mol/L sulfuric acid.The CarboPacTM PA20 column was selected for separation.5 mmol/L sodium hydroxide solution was used as the eluent for gradient elution,with the detection of pulse amperometric detector(PAD).The results showed that the six sugars could be separated completely under the above conditions,the linear relationship of glucose and other sugars within the range of 0.25~2.50μg/mL and 0.025~0.25μg/mL,respectively,was good.And the correlation coefficient was more than 0.998.The RSD of method was within 2.3%~5.5%,and the recovery was within 79.6%~116.5%.The method has the advantages of simple operation,high sensitivity and good resolution,and can be used to determine the content of sugars in beef hydrolysates accurately and rapidly.
