摘要 目的:探讨叉状头转录因子(FoxP3)/T细胞免疫球蛋白黏蛋白分子-3(Tim-3)信号通路中的相关基因在系统性硬化症(SSc)中的表达。方法:收集我院30例系统性硬化症患者和15例健康对照的外周血,分离得到外周血单核细胞(PBMCs),流式细胞术检测PBMCs中CD4+CD25+Treg细胞和Th17辅助细胞(Th17)变化,RT-PCR检测PBMCs中FoxP3和Tim-3mRNA变化,ELISA检测细胞毒T淋巴细胞相关抗原4(cytotoxic T lymphocyte-associated antigen-4,CTLA-4)及γ干扰素(IFN-γ)含量。结果:与健康对照组相比,SSc患者PBMCs中Treg细胞和Th17细胞数目上升,但二者比值下降,FoxP3mRNA表达量下降,而Tim3mRNA表达量上升;CTLA-4及IFN-γ含量皆下降。结论:SSc患者PBMCs中T调节细胞(Treg)和Th17水平提高,但功能上存在缺陷,可能与FoxP3/Tim-3信号通路中相关基因表达变化有关。 Objective: To explore the molecular of immunology of FoxP3/Tim-s signaling pathway related gene in systemic sclerosis (SSc). Methods.. Peripheral blood was collected from 30 SSc patients and 15 healthy cases. The ratio of CD4^+ CD25^+ Treg and Th17 were measured by flow cytometry. The FoxP3 and Tim 3 mRNA were detected by RT-PCR. The level of CTLA-4 and IFN-)' was determined by elisa. Results: The cell number of CD4^+ CD25^+ Treg and Thl7 from peripheral blood mononuclear cells (PBMCs) of SSc patients was higher than the healthy group, but the ratio of Treg/Th17 was lower than the healthy group. Compared with the healthy group, the expression of FoxP3 from PBMCs of SSc patients was decreased, the ex- pression of Tim-3 from PBMC of SSc patients was increased. The level of CTLA-4 and IFN y from PBMC of SSc patients was lower than the healthy group, Conclusions: The levels of Treg and Th17 from PBMCs of SSc patients are increased, but defects exist on the function, which may be associated with FoxP3/Tim 3 signaling pathway related gene changes.
