摘要
目的探讨Cerb B-2与MDR1/P-gp在乙状结肠黏液腺癌细胞表达及与临床预后资料的相关性。方法应用免疫组织化学及组织芯片技术,检测86例乙状结肠黏液腺癌组织Cerb B-2与MDR1/P-gp的表达情况。结果Cerb B-2在临床分期Ⅲ期阳性表达率为76.19%(16/21),明显高于Ⅰ、Ⅱ期;Cerb B-2在有腋窝淋巴结转移组阳性表达率为65.71%,明显高于无腋窝淋巴结转移组的表达(37.25%),差异有统计学意义(P<0.05)。MDR1/P-gp在临床Ⅲ期阳性表达率与Ⅰ、Ⅱ期相比,差异无统计学意义(P>0.05);有腋窝淋巴结转移组MDR1/P-gp的阳性表达率为62.85%,明显高于无腋窝淋巴结转移组的表达(29.41%),差异有统计学意义(P<0.05)。MDR1/P-gp蛋白阳性表达组Cerb B-2同时阳性者占72.97%(27/37),Cerb B-2表达阴性者占54.05%(20/37),两者差异有统计学意义(P<0.01)。不同肿瘤大小Cerb B-2与MDR1/P-gp的表达比较,差异无统计学意义(P>0.05)。结论 Cerb B-2与MDR1/P-gp的阳性表达在乙状结肠黏液腺癌多药耐药发生中具有一定作用,联合检测有利于对乙状结肠黏液腺癌的综合治疗提供依据。
Objective To investigate the correlation between the expression of CerbB-2 and MDR1 /P-gp in sigmoid colon mucinous adenocarcinoma and clinical prognostic data.Methods The expression levels of CerbB-2 and MDR1 /P-gp were detected by immunohistochemistry and tissue microarray technique in 86 cases of sigmoid colon mucinous adenocarcinoma tissues.Results The positive expression rate of CerbB-2 in patients at stage Ⅲ was 76.19%（16 /21）,which was significantly higher than that of patients at stage Ⅰand stage Ⅱ.The positive expression rate of CerbB-2 in patients with axillary lymph node metastasis was 65.71%,which was significantly higher than that （37.25%）of patients without axillary lymph node metastasis （P 〈0.05）.However there was no significant difference in the positive expression rate of MDR1 /P-gp between stage Ⅲ and stage Ⅰ,stage Ⅱ（P 〉0.05）.The positive expression rate of MDR1 /P-gp in patients with axillary lymph node metastasis was 62.85%,which was significantly higher than that （29.41%）of patients without axillary lymph node metastasis （P 〈0.05）.Both positive expressions of MDR1 /P-gp and CerbB-2 accounted for 72.97%（27 /37）,however,the negative expression of CerbB-2 accounted for 54.05%（20 /37）,there was significant difference between two groups （P 〈0.01 ）. However there was no significant difference between the expression of CerbB-2 and MDR1 /P-gp in different size tumors （P 〉0.05）.Conclusion The positive expression of CerbB-2 and MDR1 /P-gp plays a certain role in the pathogenesis of multidrug resistance of sigmoid colon mucinous adenocarcinoma,thus combined detection of CerbB-2 and MDR1 /P-gp is helpful to provide basis for the comprehensive treatment of sigmoid colon mucinous adenocarcinoma.
