摘要
目的:观察针刀松解法对第三腰椎(L3)横突综合征大鼠脊髓背角前阿黑皮素(POMC)、前脑啡肽原(PPE)mRNA表达的影响,探讨其中枢镇痛机制。方法:选用SD雄性大鼠,随机分为4组:正常组、模型组、电针组、针刀组,后3组文献方法建立左侧L3横突综合征的动物模型,针刀组和电针组分别给予针刀松解和电针干预。造模28d后取材,应用mRNA原位杂交方法检测大鼠脊髓背角POMC mRNA、PPE mRNA阳性细胞的表达。结果:造模后,模型组大鼠脊髓背角POMC、PPE mRNA的阳性细胞表达较正常组显著增多,阳性强度显著增高(P<0.05,P<0.01),针刀组和电针组大鼠脊髓背角POMC mRNA、PPE mRNA阳性细胞表达较模型组显著减少,阳性强度显著降低(P<0.05,P<0.01)。结论:针刀松解法或电针可能通过调解脊髓背角POMC mRNA、PPE mRNA的表达,参与镇痛过程。
Objective: To observe the efects of acupotomy lysis on mRNA expressions of POMC and PPE in spinal dorsal horn in rats with the third lumbar vertebrae transverse process syndrome and explore its analgesia mechanism in central nervous system.Methods: SD rats were randomly divided into normal control group,model group,electroacupunture(EA) group and acupotomy lysis(AL) group.Except for the normal control group,the other groups were established the third lumbar vertebrae transverse process syndrome model according to the methods in literature.AL group and EA group were respectively intervened with AL and EA.POMC mRNA and PPE mRNA positive cell contents in spinal dorsal horn were tested by situ hybridization method on day 28 after modeling.Results: In the model group,the numbers of POMC mRNA and PPE mRNA positive cells were increased in spinal dorsal horn than those in normal group,positive stain intensity was increased,as well(P〈0.05,P〈0.01).In AL group and EA group,the number of POMC mRNA,PPE mRNA positive cells and positive stain intensity were decreased than those in model group(P〈0.05,P〈0.01).Conclusion: AL and EA therapy may participate in the analgesia which may be related to modulating the expressions of POMC mRNA and PPE mRNA in spinal dorsal horn.
