脉冲电磁场与正弦交变电磁场对成骨细胞增殖与成熟矿化的比较研究 被引量:5

摘要 目的探讨0.6 m T 50 Hz脉冲电磁场和1.8 m T 50 Hz正弦交变电磁场对大鼠颅骨成骨细胞增殖与成熟矿化的影响。方法采用酶消化法分离大鼠乳鼠颅骨成骨细胞,接种培养于10%胎牛血清(FBS)的α-MEM培养基中,成骨细胞融合到80%~90%,传代培养,随机分成3组,分别为对照组、脉冲电磁场组和正弦交变电磁场组,分别测定细胞增殖、碱性磷酸酶(ALP)活性,并进行碱性磷酸酶染色和茜素红钙化结节染色。结果脉冲电磁场组OD值明显高于正弦交变电磁场组和对照组(P〈0.01),正弦交变电磁场组明显低于对照组(P〈0.01);第6天脉冲电磁场组ALP活性明显高于正弦交变电磁场组和对照组(P〈0.01),第9天脉冲电磁场组和正弦交变电磁场组ALP活性均明显高于对照组(P〈0.01)。脉冲电磁场组和正弦交变电磁场组ALP染色、茜素红面积和克隆数均显著高于对照组(P〈0.01)。结论脉冲电磁场和正弦交变电磁场都能最终促进成骨细胞的成熟矿化,使其形成新骨,但其作用机理不同。 Objective To investigate effects of 0. 6 m T 50 Hz pulsed electromagnetic fields( PEMFs) and1. 8 m T 50 Hz sinusoidal electromagnetic fields( SEMFs) on proliferation and maturity mineralization of skull osteoblasts( OB) in rats. Methods Enzyme digestion was used to obtain bone cells from neonatal SD rats' skulls,and the cells were cultured in α-MEM containing 10% fetal bovine serum( FBS). When the OB values reached 80%-90% confluence,they were given serial subcultivation and were randomly divided into control group,PEMFs group and SEMFs group. The cell proliferations,alkaline phosphatase( ALP) activity,ALP staining and calcified node staining were respectively detected in the three groups. Results The optical density( OD) value in PEMFs group was significantly higher than those in SEMFs and control groups( P〈0. 01),while the value in SEMFs group was significantly lower than that in control group( P〈0. 01); ALP activity on 6thd after treatment was significantly increased compared with those in control and SEMFs groups( P〈0. 01),and ALP activities on 9thd after treatment in PEMFs and SEMFs groups were significantly increased compared with that in control group( P〈0. 01). The values of ALP staining,Alizarin red area and cloning number in PEMFs and SEMFs groups were significantly higher than those in control group( P〈0. 01). Conclusion Both PEMFs and SEMFs can eventually promote osteoblasts proliferation and maturity mineralization,and then induce formation of new bones with different mechanisms.
