摘要
目的:研究成骨生长肽OGP在体外对大鼠颅盖骨成骨细胞I型胶原蛋白的影响。方法:在体外培养的新生大鼠颅盖骨成骨细胞中加入浓度为10-11~10-7mol/L的OGP,处理7天后收集细胞爬片,采用SABC法行细胞免疫化学染色,通过图像分析系统观察不同OGP作用后,成骨细胞胶原蛋白的变化情况。结果:OGP作用于成骨细胞后,I型胶原蛋白表达明显增强,其中OGP浓度为10-9mol/L时促进作用最为明显。结论:OGP可促进成骨细胞I型胶原蛋白的表达。
Objective:To elucidate the effect of osteogenic growth peptide (OGP) on the collagen Ⅰ in cultured rat osteoblast-like cells. Method: Osteoblast-like cells were treated with 10^-11~10^-7 mol/L of OGP for 7d respectively. Then cells were processed for SABC stain and examination of the collagen Ⅰ. Microscopy and analysis system was used to investigate the changes of collagen Ⅰ. Result:OGP can promote the expression of collagen Ⅰ in rat osteohlast-like cells. The most best concentration of OGP is 10^-9 mol/L. Conclusion:Our results demonstrate that OGP at 10^9mol/L may stimulate the expression of collagen I in rat osteoblast-like cells in vitro.
