积雪草苷对心肌梗死大鼠TGF-β1、ColⅠ和Col Ⅲ表达的影响 被引量:1

The effect of Asiaticoside on the expression of TGF-β1,ColⅠand Col Ⅲ in rats after myocardial infarction

摘要 目的研究积雪草苷对心肌梗死(心梗)大鼠左室心肌纤维化和心肌组织转化生长因子-β1(transforming growth factor-β1,TGF-β1)、胶原Ⅰ(collagenⅠ,ColⅠ)和胶原Ⅲ(collagenⅢ,ColⅢ)表达的影响。方法结扎雄性SD大鼠左冠状动脉前降支建立心梗模型,随机分为假手术组(Sham组)、假手术+积雪草苷组(Sham+Asiaticoside组)、心梗模型组(MI组)和心梗模型+积雪草苷组(MI+Asiaticoside组)。造模当天给药,早晚各1次,2周后分别测定各组大鼠心脏左室重量与体重之比、左室梗死面积;免疫组织化学(免疫组化)检测心肌组织非梗死区TGF-β1、ColⅠ、ColⅢ的表达。计量资料采用t检验,P<0.05为差异有统计学意义。结果 MI组左室梗死面积为(24.37±5.69)%,MI+Asiaticoside组左室梗死面积为(25.89±6.27)%,差异无统计学意义(P>0.05);MI组左室重/体重为(3.46±0.26)mg/g,MI+Asiaticoside组左室重/体重为(2.53±0.35)mg/g,差异有统计学意义(P<0.05);免疫组化染色结果显示,与MI组相比[(8.34±0.62)、(37.66±2.21)、(12.39±1.37)μm2],MI+Asiaticoside组非梗死区心肌TGF-β1、ColⅠ、ColⅢ的表达[(5.97±0.78)、(17.90±1.36)、(8.14±1.23)μm2]明显降低,差异均有统计学意义(均P<0.05)。心梗大鼠非梗死区心肌组织ColⅠ、ColⅢ表达与心肌组织TGF-β1的表达正相关。结论积雪草苷可减少心梗大鼠非梗死区TGF-β1的表达,从而减弱非梗死区心肌组织ColⅠ、ColⅢ异常合成与沉积,减轻心梗后左室心肌纤维化程度,延缓左室重构的发展。 Objective To study the effect of Asiaticoside on myocardial fibrosis and the expression of transforming growth fac- tor-β1(TGF-β1) and collagen Ⅰ (Col Ⅰ) and collagen Ⅲ (Col Ⅲ) of left ventricular non-infarcted myocardium in rats after myocardial infarction.Methods MI model was established in male SD rats by ligation of left anterior descending coronary artery.The rats were randomly divided into four groups:sham-operated group(Sham group),sham-operated+Asiaticoside group(Sham+Asiaticoside group),MI model group(MI group) and Asiaticoside treatment group(MI+Asiaticoside group) after myocardial infarction.After 2 weeks of treatment,the cardiac function was assessed by the ratio of left ventricular weight and body weight,infarct size.The proteins of myocardial TGF-β1 and Col I and Col m of non-infarcted myocardium were determined through immunohistochemical methods. Groups were compared using t test,P〈0.05 as statistically significant difference.Results Left ventricular infarct size of MI model group was (24.37 ± 5.69)%,and left ventricular infarct size of Asiaticoside treatment group was (25.89± 6.27)%,the difference was not statistically significant(P〉0.05).The ratio of left ventricular weight and body weight in MI model group was (3.46 ± 0.26)mg/g, and the ratio of left ventricular weight and body weight in MI+Asiaticoside treatment group was (2.53 ± 0.35)mg/g,the difference was statistically significant(P〈0.05).Immunostained TGF-131,Col I and Col Ⅲ of non-infarcted myocardium were obviously lower in the group. MI+Asiaticoside than those in the MI model group[(5.97 ± 0.78),(17.90 ± 1.36),(8.14 ±1.23),(8.34 ± 0.62),(37.66± 2.21), (12.39 ± 1.37)μm2](all P〈0.05).The expression of Col Ⅰ ,Col Ⅲ and the expression of TGF-131 in rats after myocardial infarction showed a significant positive correlation.Conclusion Asiaticoside can inhibit myocardial fibrosis through decreasing the expression of T
