摘要
目的:探讨复方积雪草含药血清对炎症介质诱导的肾小管上皮细胞表型转化的干预作用。方法:用IL-1β(10ng/ml)刺激原代小鼠肾小管上皮细胞,并用低、中、高浓度复方积雪草含药血清进行干预。应用MTT法测定复方积雪草含药血清的细胞毒性;Western-blottlng和逆转录-聚合酶链反应(RT-PCR)分别测定细胞α-SMA、Vimentin蛋白和基因表达。结果:IL-1β能够诱导肾小管上皮细胞α-SMA、Vim-entin蛋白质合成增加,基因表达亦显著上调;各个浓度的复方积雪草含药血清干预均可显著降低α-SMA、Vimentin蛋白质、基因的表达。结论:复方积雪草含药血清可抑制免疫炎症因子诱导的肾小管上皮细胞表型转化,从而在防治肾小管间质纤维化中发挥其作用。
Objective:To explore the effects of serum containing centella asiatica compound on phenotype transdifferentiation of renal tubular epithelial cells.Methods: Mice's primary tubular epithelial cells were cultured in vitro,model cells were stimulated by IL-1β,the cells in medicine groups were co-incubated with low,middle and high concentrations serum containing centella asiatica compound at the same time.The protein expression of α-SMA and Vimentin were detected by Western-blotting,α-SMA and vimentin mRNA were determined by RT-PCR.Results: The mRNA expression and protein levels of α-SMA and vimentin in renal tubular epithelial cells stimulated with IL-1β were significantly higher than those in normal,serum containing centella asiatica compound of different concentrations could significantly reduce the mRNA expression and protein levels of α-SMA and vimentin in renal tubular epithelial cells.Conclusion:Centella asiatica compound can reduce the phenotype transdifferentiation of renal tubular epithelial cells stimulated by inflammatory factor and prevent renal interstitial fibrisis.
