积雪草对高糖诱导的大鼠NRK-52E细胞CTGF和FN表达的影响 被引量:6

Effect of Centella Asiatica on Expression of Connective Tissue Growth Factor and Fibronectin in NRK52E Cells Stimulated by High Glucose

摘要 目的观察积雪草含药血清对高糖刺激下大鼠近端肾小管上皮细胞株NRK52E表达结缔组织生长因子(CTGF)及纤维连接蛋白(FN)的影响,进一步探讨积雪草抑制肾小管间质纤维化,防治糖尿病肾病的作用机制。方法体外培养NRK52E细胞分为正常对照组、模型组、积雪草高剂量组、积雪草中剂量组、积雪草低剂量组及贝那普利组6组进行处理,采用Western Blot及Real time PCR方法检测各组CTGF、FN蛋白及m RNA表达。结果与对照组比较,模型组CTGF、FN蛋白及m RNA表达明显升高(P<0.05);与模型组比较,积雪草不同剂量组及贝那普利组CTGF、FN蛋白及m RNA表达均明显下调(P<0.05),且随着积雪草剂量的增大,这种下调趋势逐渐明显。结论积雪草可能通过调控CTGF、FN蛋白及m RNA表达,抑制肾小管间质纤维化,进而防治糖尿病肾病。 Objective: To investigate the effect of centella asiatica on expression of CTGF and FN in NRK52 E cells stimulated by high glucose,and further explore the mechanism of centella asiatica on inhibiting renal tubular interstitial fibrosis and treatment of diabetic nephropathy. Methods: NRK52 E cells were cultured and divided into five groups: the normal control group,high glucose group,groups of different dose of centella asiatica and captopril group. Western blot was used to detect the expression of CTGF and FN protein,and Real time PCR was used to detect the expression of CTGF and FN m RNA. Results: Compared with the normal control group,the expression of CTGF and FN protein and m RNA of model group were up-regulated obviously(P 0.05);The expression of CTGF and FN protein and m RNA of model group down-regulated markedly when treated with different dose of centella asiatica and captopril,and the downtrend was gradually obvious with the increase of centella asiatica dose. Conclusion: Centella asiatica can inhibit renal tubular interstitial fibrosis through regulating expression of CTGF and FN in m RNA and protein levels.
