番茄红素诱导K562细胞凋亡及机制的探讨 被引量:3

摘要 目的:研究番茄红素(lycopene)对人慢性髓系白血病K562细胞凋亡的影响,探讨其凋亡的作用机制。方法:用不同浓度(0,20,40,60μmol·L-1)的番茄红素作用K562细胞24,48,72 h,0μmol·L-1番茄红素为空白组。采用MTT法检测不同浓度的番茄红素作用24,48,72 h时K562细胞增殖抑制率;Annexin V/PI双染色法检测不同浓度的番茄红素作用48 h时K562细胞凋亡率;RT-PCR法及Western blot法检测不同浓度的番茄红素作用48 h时K562细胞中B细胞淋巴瘤-2(Bcl-2),半胱氨酸天冬氨酸蛋白酶-3(Caspase-3)mRNA及蛋白的表达。结果:与空白组比较,细胞增殖抑制率随番茄红素浓度升高而明显升高,细胞凋亡率逐渐明显增加,随番茄红素浓度明显升高,Bcl-2 mRNA及蛋白的表达明显降低,Caspase-3mRNA及蛋白的表达明显升高,均具有统计学差异(P<0.05)。结论:番茄红素诱导人慢性髓系白血病K562细胞凋亡,其凋亡机制与下调Bcl-2及上调Caspase-3表达有关。 Objective: To investigate the induced apoptotic effects and its mechanism of lycopene on human leukemia cell line K562 cells. Method: K562 cells were treated with different concentrations of lycopene (0, 20, 40, 60 μmol·L^-1). Cell proliferation in K562 was observed by MTT. Cell apoptosis was inspected by Annexin V/PI double staining method. The expressions of Bcl-2 and Caspase-3 were determined using RT-PCR and Western blot. Result: MTT and Annexin V/PI showed that the rates of proliferation inhibition and apoptosis were positively associated with the concentrations of lycopene (P 〈 0.05). RT-PCR and Western blot indicated that the expression of Bcl-2 gene decreased negatively, but the expression of Caspase-3 gene decreased positively associated with concentration of lycopene (P 〈 0.05). Conclusion: Lycopene could induce the apoptosis of K562 cells, which may be associated with lowering expressions of Bcl-2 and enhancing caspase-3 activity.
