摘要
目的分析肝癌组织中乙型肝炎病毒(HBV)感染患者体内DNA甲基转移酶(DNMT)表达量,探讨肝癌组织中HBV感染与甲基化转移酶表达的关系。方法选取2013年1月间至2014年1月青岛市中心医院放疗科接受治疗的52例肝癌患者,根据肝癌组织中HBV感染情况分为HBV感染组和非HBV感染组,每组26例。采用荧光定量聚合酶链式反应(PCR)方法,检测52例患者肝癌组织内DNMT1、DNMT2、DNMT3a和DNMT3b的mRNA表达量。结果 HBV感染组DNMT1 mRNA表达为1.21±0.24,非HBV感染组为0.83±0.21,HBV感染组DNMT1 mRNA表达高于非HBV感染组,差异有统计学意义(t=6.08,P<0.05);HBV感染组DNMT2 mRNA表达为0.77±0.15,非HBV感染组为0.63±0.12,HBV感染组DNMT2mRNA表达高于非HBV感染组,差异有统计学意义(t=3.72,P<0.05);HBV感染组DNMT3a mRNA表达为1.36±0.25,非HBV感染组为0.94±0.17,HBV感染组DNMT3a mRNA表达高于非HBV感染组,差异有统计学意义(t=7.08,P<0.05);HBV感染组DNMT3b mRNA表达为0.68±0.12,0.49±0.09,HBV感染组DNMT3b mRNA表达高于非HBV感染组,差异有统计学意义(t=6.46,P<0.05)。结论肝癌组织中HBV感染促进甲基化转移酶的表达,从而促进抑癌基因甲基化。
Objective To analyze the in vivo expression of methyl transferee's enzyme in hepatocellular carcinoma( HCC) patients with / without hepatitis B virus( HBV) infection and discussion the relationship between HBV infection and hepatocellular carcinoma methylamine transferees expression. Methods Between January 2013 and January 2014,52 patients with HCC treated in Qingdao Central Hospital were enrolled in the study. According to with HBV infection or not,the patients were divided into two groups: HBV infection group and non-HBV infection group,with 26 cases in each group. DNA methyltransferase( DNMT) 1,DNMT2,DNMT3 a and DNMT3 b mRNA expressions were evaluated using fluorescence quantitative polymerase chain reaction( PCR). Conclusion HBV infection promotes the expression methyltransferase enzyme,thereby promoting tumor suppressor gene methylamine.
