阿司匹林抵抗与环氧合酶1基因多态性关联研究 被引量:2

Correlation between COX-1 gene polymorphism and aspirin resistance

摘要 目的探讨心脑血管病人群阿司匹林抵抗(aspirin resistance,AR)与环氧合酶1(cycloxygenase,COX-1)基因4个单核苷酸多态性(Single Nucleotide Polymorphisms,SNP)位点的相关性。方法采用引物3'端错配扩增阻滞(ARMS)原理,结合荧光定量PCR技术(TaqMan探针)对经颅脑MRI及冠脉造影证实的380例缺血性卒中(Ischemic Stroke,IS)及冠心病(coronary artery disease,CAD)患者进行分组检测。结果 COX-1的22C>T、50C>T位点未见多态性,-842A>G基因多态位点3种基因型(AA、A/G、GG)在AS组、ASR与AR组分布频率分别为70.3%,21.5%,8.2%;100%,0%,0%;94.1%,0%,3.7%,3种基因型在3组间的分布差异无统计学意义(P=0.060)。128G>A基因多态位点3种基因型(GG、G/A、AA)在AS组、ASR与AR组分布频率分别为72.0%,20.3%,21.5%,7.7%;70.2%,19.1%,10.6%;50.0%,,50.0%,0%,3种基因型在3组间的分布差异无统计学意义(P=0.800)。两基因单体型构建在3组间无统计学意义。结论 COX-1的-842A>G、128G>A位点基因多态性与AR发生无关联。 Objective To study the correlation between single nucleotide polymorphism at-842A〉G,22C〉T,50C〉T,128G〉A of cyclooxygenase-1(COX-1) gene and aspirin resistance(AR) in patients with cardiovascular and cerebrovascular diseases.Methods According to the rate of platelet aggregation,380 patients with cardiovascular and cerebrovascular diseases were divided into AR group(31 cases),aspirin semi-resistance(ASR) group(71 cases) and aspirin sensitivity(AS)group(268 cases).The SNPs at-842A〉G,22C〉T,50C 〉T,128G〉A of cyclooxygenase-1(COX-1) gene were detected in these cases using amplification refractory mutation system(ARMS) in combination with TaqMan probe-based fluorescent quantitative PCR,then genotypes and alleles distribution of polymorphism locus of COX-1 gene were analyzed among the three groups.And haplotypes were built as well.Results The genotype distribution frequencies of COX-1-842A〉G genotypes(GG,GA,AA) in the AS group,ASR group and AR group(92.9%,0.4%,6.7%;100%,0%,0%;96.8%,0%,3.2%;respectively) had no significant difference(P = 0.246).There was also no significant difference of the genotype frequencies of COX-1-128G〉A among the three groups(86.2%,13.4%,0.4%;84.6%,14.0%,1.4%;80.6%,19.4%,0%;P = 0.800).Both haplotypes built by COX-1-842A〉G and 128G〉A had no significant difference among the three groups too(P = 0.077,0.696,0;0.869,0.396,0.563;respectively).Conclusion There was no significant correlation between single nucleotide polymorphism at-842A〉G,22C〉T,50C〉T,128G A of COX-1 gene and AR in patients with cardiovascular and cerebrovascular disease.
