摘要
目的探讨亚低温对急性肺损伤(Au)大鼠Toll样受体2(TLR2)蛋白表达及下游促炎性因子的影响。方法按随机数字表法将90只雄性SD大鼠分为四组:亚低温组、控温组、非控温组[气管滴入脂多糖(LPS)制备ALI大鼠模型]及对照组(滴入等量的生理盐水)。建模成功后,亚低温组及控温组分别将体温维持在33~34℃、36—37℃。分别于滴入LPS或生理盐水后2h、7h及13h处死大鼠。分别于术前及术后1h、2h、7h、13h测定动脉血气分析。光镜下观察肺组织形态结构,进行病理评分。分别采用蛋白质印迹法(Westernblot)及逆转录-聚合酶链反应(RT-PCR)检测大鼠肺组织的TLR2蛋白表达及髓样分化因子(MyD88)、肿瘤坏死因子-α(TNF-α)、诱导型一氧化氮合酶(iNOS)mRNA的转录。结果LPS干预后,肺组织病理评分升高;TLR2蛋白及Myd88、TNF-α、iNOSmRNA表达随时间延长呈升高趋势(均P〈0.01)。1h、6h亚低温组氧分压好于同时段控温组及非控温组(均P〈0.05),12h亚低温组氧分压与同时段控温组及非控温组比较差异无统计学意义(均P〉0.05)。同一时段亚低温组肺组织评分低于控温组及非控温组(均P〈0.05),TLR2蛋白表达低于控温组及非控温组(均P〈0.01),MyD88mRNA(2^-△△CT)低于控温组及非控温组(均P〈0.01),TNF—α mRNA(2^-△△CT)低于控温组及非控温组(P〈0.01),iNOSmRNA(2^-△△CT)低于控温组及非控温组(均P〈0.01)。结论亚低温能降低内毒素诱导的ALI大鼠的TLR2蛋白及下游促炎症因子表达,减轻肺损伤。
Objective To investigate the impact of hypothermia (HT) on TLR2 protein expression and downstream inflammatory mediators during acute lung injury (ALI). Methods Ninety male SD rats were randomly divided into four groups. The ALI model was reproduced by LPS intratracheal instillation ( hypothermia group, HT ; temperature - controlled group, TC ; temperature - uncontrolled group, UTC) ; saline was instilled intratracheally for control group. In HT group and control temperature group the body temperature was respectively maintained to 33 - 34 ℃ and 36 -37 ℃ 1 hour after LPS instillation. All the subject were sacrificed after LPS or saline instiUation for 2 h, 7 h, 13 h. The morphological changes in lung tissue of rats were observed under light microscope. TLR2 protein expression, myeloid differentiation factor88 ( Myd88 ), tumour necrosis factor - α ( TNF - α) and inducible nitric oxide synthase (iNOS) mRNA transcriptional level was determined by western blot andreverse transcription- polymerase chain reaction (RT- PCR). Results With the intervention of LPS, oxygen partial pressure of each group was decreased obviously; score of morphological changes in lung tissue was increased significantly TLR2 protein expression, Myd88, TNF -α and iNOS mRNA transcription in HT group, TC group and UTC group was in exhibited an increased trend (both P 〈 0. 01 ). At 1 hour and 6 hour, oxygen partial pressure in hypothermia group was higher than that in TC group and UTC group ( both P 〈 0.05 ) ; at 12 hour, oxygen partial pressure in HT group and UTC group showed no difference ( both P 〉 0. 05). At the same time, score of morphological changes in lung tissue in hypothermia group was lower than that in TC group and UTC group ( both P 〈 0.01 ) ; TLR2 protein expression in hypothermia group was lower than that in TC group and UTC group ( both P 〈 0.01 ) ; Myd88 mRNA expression (2^-△△CT) in hypothermia group was lower than that in TC group and UTC group (both P �
