姜黄素调控结肠癌SW480细胞skp2-p27kip通路的研究 被引量:4

摘要 目的:探讨姜黄素抑制结肠癌细胞增殖并促进其凋亡的分子机制。方法:通过MTT检测不同浓度姜黄素(0,7.5,15,30,60μmol/L)处理结肠癌细胞SW480后凋亡情况;通过蛋白免疫印迹方法检测不同浓度姜黄素(0,7.5,15,30,60μmol/L)处理结肠癌细胞后细胞中skp2、p27kip蛋白表达情况。结果:姜黄素能抑制结肠癌细胞SW480增殖并促进其凋亡,有浓度和时间依赖;姜黄素降低了结肠癌细胞SW480中skp2蛋白的表达(P<0.05),而促进了p27kip蛋白的表达,有时间和剂量依赖,差异有统计学意义(P<0.05)。结论:姜黄素能抑制结肠癌细胞SW480的增殖并促进其凋亡,有时间和剂量依赖,这种抗癌效应可能与干扰skp2-p27kip通路有关。 Objective: To detect the mechanism of curcumin inhibiting the proliferation and improving the apoptosis of col-orectal cancer cells. Methods: The study tested the apoptosis of colorectal cancer cells caused by different concentrations of cur-cumin(0, 7.5, 15, 30, 60 μmol/L) with MTT counting method, and observed the protein expression of skp2 and p27 kip in SW480 which were treated with different concentrations of curcumin(0, 7.5, 15, 30, 60 μmol/L) with western blot assay. Results:Curcumin could suppress the proliferation activity of colorectal cancer cell SW480 and accelerate its apoptosis, with time-dose dependence. Curcumin could decrease the protein expression of skp2(P〈0.05), and promote the protein expression of p27 kipof colorectal cancer cell SW480(P〈0.05), with time-dose dependence. Conclusion: Curcumin could inhibit the proliferation activity and accelerate the apoptosis of colon cancer cells SW480, with time-dose dependence, and the effect of anticancer may be as-sociated with the interference skp2-p27 kip pathway.
