摘要
为了选育高纤维素酶活的Trichoderma asperelloides菌株,本试验以中国工业微生物菌种保藏管理中心的Trichoderma asperelloides41245为原始菌株,采用紫外和微波两种物理方法复合诱变,纤维素刚果红平板初筛和固态发酵产酶复筛。试验最终获得一株Trichoderma asperelloides突变株ZWWB1,在以麦秸和麸皮为基质的固态发酵培养基上培养96 h,滤纸酶活力达15.08 U/g,是原始菌株的1.81倍。经5次传代发酵培养滤纸酶活力变化不大。可见该突变菌株ZWWB1产纤维素酶能力较高且遗传稳定性较好,可考虑进一步开发应用于秸秆饲料发酵。
The aim of this study was to screen Trichoderma asperelloides strain with high cellulase activity.Trichoderma asperelloides41245 of China Center of Industrial Culture Collection was used as original strain in this experiment. Compound mutation by ultraviolet(UV)and microwave irradiation, primary screening by cellulose-congo red plate and secondary screening by solid state fermentation was adopted.A mutant strain of Trichoderma asperelloides ZWWB1 was obtained ,and its filter paper activity(FPA)reaehed 15.08 U/g after fermented for 96 hours with wheat straw and bran as solid matrix,which was 1.S1 times the activity of the original strain.After transferring for five generations and culturing in solid, its FPA was a little changed.This mutant strain ZWWB1 has high cellulase-producing activity and better hereditary stabil- ity ,which could be further developed and applied in straw feed fermentation.
